| Objective Hepatocellular carcinoma(HCC)is one of the most common primary liver cancers,and it has the characteristics of high morbidity and mortality.Although there are many ways to treat HCC such as surgery,ablation,chemotherapy and targeted drug therapy,there is still a high recurrence rate.In recent years,the research on the regulation between various signaling pathways and HCC has attracted much attention,and about 50%of HCCpatients are related to PI3K/AKT signaling pathway.Therefore,this study will focus on the mechanism of PI3K/AK signaling pathway in HCC.Wogonoside is one of the active components extracted from Scutellaria baicalensis Georgi.At present,there are few reports about the anti-tumor effect of wogonin in liver cancer at home and abroad.This study intends to explore the correlation between wogonin and PI3K/AKT signaling pathway in HCC through in vitro cell experiments,so as to clarify the therapeutic mechanism of wogonoside on HCC.Methods 1.Using the CCK-8 method to detect the activity of different doses of wogonoside treatment of L-O2 cells of normal liver cells and liver cancer HepG2 cells(the concentrations of wogonoside drugs are 0 μM,50 μM,100 μM,200 μM,250 500 750 μM,1000μM)after 24h,48h and 72h.2.Using EDU and clone formation assay to detect the proliferation ability of liver cancer HepG2 cells.3.The migration and invasion ability of liver cancer HepG2 cells were detected by Transwell assay and scratch assay.4.The apoptosis of liver cancer HepG2 cells was detected by flow cytometry Annexin V-FITC/PI double staining method.5.RT-PCR and Western blot were used to detect the expression changes of apoptosis-related proteins Bax,Bcl-2 and PI3K/AKT signaling pathway proteins.Results 1.Compared with the blank control group,the activity,proliferation,invasion,migration and healing ability of hepatoma HepG2 cells decreased significantly with the increase of the concentration of wogonoside and the extension of intervention time,and the difference was statistically significant(P<0.01).Compared with the control group,wogonoside promoted the apoptosis of hepatocellular carcinoma HepG2,and the higher the concentration,the more obvious it was,with statistical significance(P<0.01).2.Compared with the blank control group,wogonoside can significantly reduce the expression level of Bcl-2/Bax relative protein,especially in the high-dose group,with statistical significance(P<0.01).Wogonoside also reduces the expression levels of p-PI3K/PI3K and p-AKT/AKT relative protein,especially p-PI3K/PI3K,with statistical significance(P<0.01).Compared with the blank control group,the treatment with wogonoside significantly decreased the mRNA expression of PI3K and AKT in HepG2 cells.With the increase of wogonoside concentration,the relative expression level of mRNA decreased significantly,with statistical significance(P<0.01).Conclusion Wogonoside can reduce the proliferation of HepG2 cells and promote the apoptosis of HepG2 cells,which may be related to PI3K/AKT signaling pathway. |
