Objective: To detect the expression level of LZTR1 protein in human pancreatic cancer tissues and adjacent tissues and analyze its effect on prognosis of patients with pancreatic cancer.To explore the effect and mechanism of LZTR1 protein on the proliferation and invasion of pancreatic cancer cells.Methods: The expression of LZTR1 protein in human pancreatic and paracancer tissues was detected by Western blot assay.Lentivirus was transfected into human pancreatic cancer cells to form stable overexpressed LZTR1 protein cell lines.Fluorescence microscopy was used to observe the expression of fluorescent protein in the cells.q RT-PCR and Western blot were used to verify the success of transfection from gene level and protein level respectively.CCK8 assay,Transwell assay and scratch assay were used to explore the changes of cell proliferation,migration and invasion ability after overexpression of LZTR1 protein in pancreatic cancer cell lines.Western blot was used to detect the expression levels of ERK1/2 and p-ERK1/2,and to study the pathway mechanism of the effect of LZTR1 protein on the proliferation,migration and invasion of pancreatic cancer cells.Results: Westernblot detection showed that the expression of LZTR1 protein in pancreatic cancer patients was lower than that in adjacent tissues,and the patients with low expression of LZTR1 protein had a poor prognosis.Overexpression of LZTR1 protein inhibited the proliferation,migration,invasion and other life activities of pancreatic cancer cells.Westernblot detection of pancreatic cancer cells with overexpression of LZTR1 protein showed that the content of ERK1/2 had no significant change compared with negative control group and blank control group,but the content of phosphorylated ERK1/2 decreased.Conclusion: LZTR1 protein in pancreatic cancer tissue is decreased compared with paracancer tissue,and low expression of LZTR1 protein has poor prognosis.Overexpression of LZTR1 protein can inhibit the proliferation,migration and invasion of pancreatic cancer cells,and may inhibit the proliferation,migration and invasion of pancreatic cancer cells by decreasing the phosphorylation level of ERK1/2.
