Objective:To investigate the protective effect of Taraxasterol(TS)on MPP~+induced SH-SY5Y cells by inducing TFEB mediated autophagy.Methods:MTT method was used to detect the toxic effects of different concentrations of MPP~+and dandelion sterol on SH-SY5Y cells,and appropriate MPP~+concentrations were selected to construct a PD model and appropriate concentration gradients of dandelion sterol.MTT assay was used to detect the protective effect of different concentrations of dandelion sterol on MPP~+induced SH-SY5Y cell death.PD cell models pretreated with different concentrations of dandelion sterol were used to extract total proteins.Western blotting was used to detect the expression of autophagy related proteins Beclin1,p62,and LC3II/I.The effect of dandelion sterol on the nuclear translocation of TFEB was detected by Western blotting after isolation of cytoplasmic nuclear proteins.Results:MTT results showed that MPP~+significantly decreased the cell viability of SH-SY5Y cells compared to the control group.With the increase of MPP~+concentration,cell viability decreased in a dose depen-dent manner.After 24 hours of treatment with 1.5 m M MPP~+,the cell survival rate was approximately 60%.The safe doses of dandelion sterol on SH-SY5Y cells are 1.25μg/ml,2.5μg/ml,and 5μg/ml,and it has a protective effect on MPP~+induced SH-SY5Y cell death.Western blotting results showed that dandelion sterol significantly increased the expression of autophagy related proteins LC3II and Beclin1,and decreased the expression of p62 protein.In addition,dandelion sterol can reverse the decrease in nuclear translocation of transcription factor TFEB induced by MPP~+in SH-SY5Y cells,and promote the nuclear translocati-on of TFEB.Conclusion:Taraxasterol can promote autophagy by promoting TFEB nuclear translocation,improve MPP~+induced autophagy disorder,and play a protective role on cells. |
