Objective:By detecting the expression of mucin-4(MUC4)in Temporal lobe epilepsy(TLE)patients and chronic epilepsy rat models,and comparing with normal brain tissue to find differences,the role of MUC4 in epilepsy was further explored.It provides a new therapeutic target for the treatment of epilepsy.Methods:1.In the brain tissue specimen bank of patients undergoing temporal lobe epilepsy surgery in the Affiliated Hospital of Jining Medical College,10 case specimens were randomly selected as the epilepsy group(TLE,n=10),and 3 brain tissue specimens of patients undergoing craniocerebral trauma surgery in the Affiliated Hospital of Jining Medical College were selected as the control group(CON,n=3).Immunohistochemistry of MUC4 in the brain tissues of epileptic patients was studied by quantitative fluorescence PCR(q PCR),Western blotting(WB)and immunohistochemistry(IHC).2.Thirty healthy adult male SD rats with body weight ranging from 210-230 g were selected and randomly divided into experimental group and control group.SD rats in the experimental group were given intraperitoneal injection of pilocarpine to induce epileptic state,which became an animal model of chronic epilepsy after feeding for one month(EP,n=20),while rats in the control group were injected with the same amount of normal saline into the abdominal cavity and fed under the same conditions for one month(con,n=10).The expression level of MUC4 in the hippocampus and cortex of rats in the experimental group and control group was observed by q PCR、WB and IHC.Results:1.The results of q PCR showed that the expression of MUC4 gene in TLE group was significantly higher than that in CON group(P<0.001,n=10);The expression of MUC4 gene in EP group was significantly higher than that in con group(P<0.001,n=20).2.WB showed that,compared with CON group,TLE group had significantly higher expression of MUC4 protein in hippocampus and cortex(P< 0.001,n=10);In chronic rat epilepsy model,the expression of MUC4 protein in the hippocampus and cortex of EP group was significantly higher than that of con group(P<0.001,n=10).3.Immunohistochemical results showed that the protein level of MUC4 was increased in TLE group and EP group.Conclusion:1.The expression of MUC4 in the brain tissue of patients with temporal lobe epilepsy was increased.2.The expression of MUC4 in hippocampus and cortex of chronic rat epilepsy model(induced by lithium chloride and pilocarpine)was increased.3.MUC4 is highly expressed in epilepsy and mainly expressed in neuron cells,which may be involved in the occurrence and development of epilepsy diseases and provide a new target for epilepsy treatment. |
