Research purpose:Fructus Ligustri Lucidi is the fruit of Ligustrum Lucidum,which has increased bone density,anti-inflammatory and liver protection properties.However,there are relatively few reports on the antibacterial activity of Fructus ligustrum.In order to further explore the residual value of Fructus Ligustrum and fill in the research blank at home and abroad.The antibacterial activity and formulation of essential oil from Fructus Ligustri Lucidi.were studied in this paper.Research method:1.Study on optimization of supercritical carbon dioxide extraction process and antibacterial activity of Fructus Ligustri Lucidi.In this experiment,a supercritical fluid extraction instrument was used to extract the essential oil from Fructus Ligustri Lucidi.and response surface was used to optimize the process.The optimal extraction process was as follows:extraction pressure 16 MPa,extraction temperature 40℃,extraction time 60 min.Under these conditions,the extraction rate was 6.21%.The preliminary bacteriostatic test was carried out by AGAR dilution method.It was found that the supercritical extract of Ligustrum Ligustrum had certain inhibitory effect on Escherichia coli,Staphylococcus aureus,Salmonella and Methicillin-resistant Staphylococcus aureus.In addition,the biosafety of Fructus Ligustri Lucidi oil was evaluated by its effects on hepatocyte proliferation and apoptosis.2.Study on the process optimization and antibacterial activity of zein membrane loaded with Fructus Ligustri Lucidi.essential oil.In this study,zein was used as film forming material and modified by adding PEG400 and glucose to improve its lack of toughness and easy fracture.Orthogonal experiment was used to optimize the process,and the optimal formula was obtained as PEG400 concentration 10%,glucose concentration 10%and drug loading 15%.After the process optimization,the maximum tension and maximum elongation of the film were adjusted in the appropriate range.The maximum tension and maximum elongation of the film were 17.43±0.60 and 15.45±0.58 respectively.Scanning electron microscopy was used to observe the cross section of the drug-loaded film,and infrared spectroscopy was used to detect whether the drug was successfully loaded.Finally,the antibacterial zone size of the drug-loaded film was measured,and the antibacterial activity of the drug-loaded film was evaluated.3.Optimization of technology and antibacterial activity of sodium alginate-gelatin composite hydrogel loaded with Fructus Ligustri Lucidi.essential oil.In this paper,a kind of gelatin-sodium alginate composite hydrogel loaded with Fructus Fructus Ligustri Lucidi.essential oil was prepared,and the process was optimized by response surface method.The drug loading,emulsifier content and CaCl2 concentration were taken as independent variables,and the encapsulation efficiency was taken as the response value.The optimum process is as follows:the content of Fructus Ligustri Lucidi.essential oil is 9.97%,the content of TW20 is 5.12%,and the concentration of CaCl2 is 1.97%.According to the actual operation and needs,the final preparation formula is:Fructus Ligustri Lucidi.essential oil content 10%,TW20 content 5%,CaCl2 concentration 2%.The swelling behavior was characterized.In addition,the drug loading was tested by Fourier transform infrared spectroscopy and scanning electron microscope.At the same time,the antioxidant and antibacterial effects of sodium alginate composite hydrogel on Staphylococcus aureus and Escherichia coli were further discussed.Research significance:In this paper,the extraction of essential oil from Ligustrum lucidum and the development of dosage forms were studied.Using environmentally friendly extraction technology and preparation materials,the dosage forms with good mechanical properties and good antibacterial activity were obtained under the premise of safety and green.This laid a foundation for the research and application of Ligustrum lucidum in antibacterial direction.
