Objective: Investigate the effects of a hyperglycemic environment on trophoblastic cell proliferation,migration,and target gene and protein expression,and to observe whether the effective extract of asiaticoside,asiaticoside glycosides,can alleviate the damage of trophoblast cells in high glucose environment,and to provide theoretical evidence for the clinical use of asiaticoside in the treatment of gestational diabetes.Methods: HTR-8/SVneo cells were cultured with culture medium containing different glucose concentrations and divided into control,high glucose and high glucose add asiaticoside groups.The differences in the expression of PI3 K,AKT2,m TORC1 and GLUT4 genes in HTR-8/SVneo cells were determined by RT-q PCR;the differences in the expression of PI3 K,AKT2,m TORC1 and GLUT4 proteins in HTR-8/SVneo cells were determined by Western-blot.Results:The results of CCK8 study showed that: high glucose environment inhibits the proliferation of HTR-8/SVneo cells,and asiaticoside promotes the proliferation of cells in high glucose environment;Scratch assay showed that:high glucose environment inhibits the migration ability of HTR-8/SVneo cells,and asiaticoside promotes the migration of cells in high glucose environment;RT-q PCR assay showed that: high glucose environment inhibited the expression levels of PI3 K,AKT2,m TORC1,GLUT4 genes in HTR-8/SVneo cells,and asiaticoside promoted the expression levels of target genes in cells compared with the high glucose group;Western-blot assay experiments showed that high glucose environment inhibited the expression levels of PI3 K,AKT2,m TORC1,GLUT4 protein expression,and asiaticoside increased targeted protein expression in HTR-8/SVneo cells.Conclusion: 1.The high glucose environment was able to suppress the proliferation and migration of HTR-8/SVneo cells and the expression of the PI3K-Akt pathway in HTR-8/SVneo cells;2.In the concentration and time range of the effect of asiaticoside measured in this experiment,asiaticoside could promote the proliferation of HTR-8/SVneo cells,and HTR-8/SVneo cells had a concentration-and time-dependent effect on asiaticoside;3.Asiaticoside can promote the expression of PI3K-Akt signaling pathway in HTR-8/SVneo cells,thereby alleviating high glucose-induced damage in HTR-8/SVneo cells..
