Objective: With the economic growth and the change of diet structure,obesity has become an important factor threatening people’s health.At present,it is found that obesity is related to a variety of diseases,and is directly related to the occurrence and development of a variety of tumors.It has been found that adipose mesenchymal stem cells(ADSC)in adipose tissue of obese patients are related to the progression of a variety of tumors.ADSC can produce a variety of cytokines,which can affect the occurrence and development of tumors.In recent years,the incidence rate of thyroid cancer has also increased continuously.The most common pathological type is thyroid papillary carcinoma,accounting for 85% of the total thyroid cancer.It has become a common endocrine malignancy.At the same time,clinical studies have found that the prevalence of thyroid papillary carcinoma in obese patients is higher than that in normal weight patients,and the tumor size,invasiveness and cervical lymph node metastasis rate in obese patients are significantly higher than those in normal weight patients,which shows that the prognosis of obese patients with thyroid papillary carcinoma is worse than that in normal weight patients.Therefore,this article will explore the effect of leptin produced by ADSC on thyroid papillary carcinoma cells in vitro.Methods: ADSCs were cultured and conditioned medium(ADSC CMS)was extracted.The concentration of leptin in conditioned medium was measured by ELISA;Then the thyroid papillary carcinoma cell line TPC-1 was intervened with ADSC CMS.The changes of TPC-1 cell viability,proliferation and migration ability under the action of ADSC CMS were detected by CCK-8,edu,Transwell,wound healing and Western blot,and its target was detected by Western blot.Then add leptin neutralizing antibody to ADSC CMS,neutralize leptin in ADSC CMS and repeat the above test to verify the effect of leptin on thyroid papillary carcinoma.Patients in the Department of thyroid surgery of the Affiliated Hospital of Southwest Medical University were selected.Thyroid papillary carcinoma tissues and normal tissues far away from the tumor were obtained during the operation.The expression of leptin receptor in thyroid papillary carcinoma tissues and normal tissues was detected by fluorescence staining.Results: Immunofluorescence detection of thyroid papillary carcinoma and normal thyroid tissues showed that there was no significant difference in the expression of leptin receptor between normal weight patients and normal thyroid tissues;Adipose mesenchymal stem cells(ADSCs)were cultured to70-80% and cultured in serum-free medium for one week to extract conditioned medium(ADSC CMS).ELISA detection of conditioned medium showed that the leptin concentration of ADSC CMS was higher than that of the control group.After the intervention of conditioned medium culture,Western blot showed that the expression level of leptin receptor in thyroid papillary cancer cells after ADSC CMS intervention was also significantly higher than that in routine culture;CCK-8,edu,Transwell and scratch tests were performed on thyroid cancer cells 24 hours after ADSC CMS intervention.It was found that ADSC CMS up-regulated the expression of matrix metalloproteinase 2(MMP2)in thyroid papillary cancer cells and promoted the proliferation and migration of thyroid papillary cancer cells to a certain extent;ADSC CMS significantly promoted the migration of thyroid cancer cell line(TPC-1)than the proliferation.After antagonizing the effect of leptin with leptin neutralizing antibody,the migration promotion decreased significantly.The above results were statistically significant.Conclusion: Immunofluorescence detection of thyroid papillary carcinoma and normal thyroid tissues showed that there was no significant difference in the expression of leptin receptor between normal weight patients and normal thyroid tissues.ADSCs up regulate the expression of leptin receptor of TPC-1 by paracrine leptin and increase the production of matrix metalloproteinase MMP-2,so as to promote the migration and proliferation of TPC-1 cells;After neutralizing leptin in conditioned medium,the promoting effect of ADSC CMS on TPC-1 migration and proliferation was weakened.This shows that leptin produced by ADSCs plays an important role in the development of thyroid papillary carcinoma.Weight control may be one of the ways to improve the prognosis of patients with thyroid papillary carcinoma. |
