Objective: This study aims to investigate the effects of Danggui Yinzi on the immune indicators of blood deficiency and wind-dry type eczema model rat and explore its mechanism,in order to provide laboratory data support for the treatment of blood deficiency and wind-dry type eczema by Danggui Yinzi.Methods: Fifty 3-week-old SD rats,half male and half female,were randomly divided into five groups: the normal group,the model group,the high-dose Danggui Yinzi group(high-dose group),the low-dose Danggui Yinzi group(low-dose group)and the desloratadine group(western medicine group).Except for the normal group,the other 4 groups used 2,4-dinitrochlorobenzene(DNCB)to make eczema model,acetylphenylhydrazine(Amphenol)combined with physical stimulation to make the model of blood deficiency and wind-dry type.After the modeling,0.9%Na Cl solution was intragastrically administered to the normal group and the model group,and the high-dose group were administrated with the crude drug concentration of 1.57 g / ml Danggui Yinzi decoction.The low dose group was treated with 0.8g / ml Danggui Yinzi decoction and the western medicine group was given 0.5mg / ml desloratadine solution for 15 days.After the intervention,the appearance,spirit,activity,skin lesions,body weight and the color of ears,claws and tail were observed.The pathological changes of skin were observed by HE staining.Serum levels of IFN-γ,IL-4,IL-18,IL-33,Ig E,LTB4,LTC4 were detected by ELISA.RT-PCR method was used to detect the expression levels of Cys LTR1 m RNA and Cys TLR2 m RNA in back skin lesions of young rats.Results:1.This experiment successfully replicated the blood deficiency wind-dry type eczema model,compared with the normal group,the model group had significantly reduced body weight(P<0.05),significantly increased spleen index(P<0.05),and significantly decreased thymus index(P<0.05),and significantly increased skin lesion score(P<0.05),skin lesions showed erythema,edema,scaling,skin thickening,lichenoid change and so on.The levels of IL-4,IL-18,IL-33,Ig E,LTB4 and LTC4 in serum of model group were significantly increased(P<0.05),however,the levels of IFN-γ were decreased(P<0.05).The levels of Cys LTR1 m RNA and Cys LTR2 m RNA in skin tissue were significantly increased(P<0.05).2.After the drug intervention,the skin lesion scores of the high and low dose Danggui Yinzi group and the western medicine group were significantly decreased(P<0.05),and the body weight and thymus index of the young rats were significantly increased(P<0.05),while the spleen index was significantly decreased(P<0.05),with the highest improvement in the high-dose Danggui Yinzi group(P<0.05).While the low-dose group of Danggui Yinzi and the western medicine group were compared,the difference was not statistically significant(P>0.05).3.The levels of IL-4,IL-18,IL-33 and Ig E in the high-dose,low-dose and western medicine groups of Danggui Yinzi were significantly decreased(P<0.05),among which the high-dose group of Danggui Yinzi was the most significant(P<0.05),while there was no statistically significant difference between the low-dose group of Danggui Yinzi and the western medicine group(P>0.05).The IFN-γ levels of the high-dose,low-dose and western medicine groups of Danggui Yinzi were significantly increased(P<0.05),with the highest increase in the high-dose group(P<0.05),while the low-dose group of Danggui Yinzi and the western medicine group had no significant difference(P>0.05).4.Danggui Yinzi could reduce the level of LTB4 and LTC4 in serum and the expression of Cys LTR1 m RNA and Cys LTR2 m RNA in skin tissue of eczema rat model of blood deficiency and wind-dry type(P<0.05),and the high dose group was better than the low dose group and western medicine group(P<0.05).Conclusion:1.Through the combined physical stimulation of DNCB and APH,the blood deficiency and wind-dry type of eczema model can be successfully replicated.The skin lesions of the model of blood deficiency and wind-dry type eczema in young rats are manifested as red spots,swelling,scales,skin thickening,moss-like changes,accompanied by weight loss of young rats,increased spleen index,reduced thymus index,increased levels of inflammatory factors in the body,and imbalance of immune system and imbalance of Th1/Th2 balance.2.Danggui Yinzi can obviously improve the appearance of skin lesions and pathological changes of blood deficiency and wind-dry type eczema in young rats,promote the weight gain of young rats,and improve the symptoms of anemia caused by acetophenone.3.Danggui Yinzi can reduce the level of inflammatory factors,correct the immune imbalance mainly caused by the hyperfunction of Th2 cells,and restore the balance of Th1/Th2 cells.4.Danggui Yinzi can inhibit the expression of inflammatory mediators and their receptors to a certain extent,thus blocking the occurrence of inflammatory response,which may be one of the mechanis of its treatment of blood deficiency and wind-dry type eczema.
