objective:Based on the observation of the clinical efficacy of enteral ointment in the treatment of intestinal mucositis and spleen and stomach qi deficiency after chemotherapy,this study further explored the protective effect of enteropathic on 5-FU induction of intestinal mucosa in rats,and conducted a preliminary study on its mechanism of action.method:1.Observation of the clinical efficacy of enterointing ointment on intestinal mucositis after chemotherapyFor 70 people who went to the Affiliated Hospital of Chuanbei Medical College for chemotherapy from July 2021 to December 2021 Patients with gastrointestinal reactions after chemotherapy were treated and observed for 8 days.The random number table method was used to divide the enrolled patients into treatment groups and control groups,with 35 cases in each group.The treatment group was given intestinal cream for external treatment,and the control group was given Western medicine for symptomatic treatment.The scores of TCM symptoms,TCM symptom efficacy,gastrointestinal symptoms and quality of life scores before and after treatment of the two groups of patients were recorded.The trial data of all enrolled patients were managed and analyzed by SPSS21.00 statistical software.2.Study on the protective effect and mechanism of enteropathic ointment on 5-FU induced rats with chemotherapy intestinal mucositis(1)Establishment of a model of 5-FU induction of chemotherapy enteroconitis in ratsRats with continuous intraperitoneal injection of 5-FU(50 mg/kg)for 5 days were observed daily for changes in weight,diarrhea and feeding volume,and changes in the pathological morphology of small intestinal tissue.(2)Protective effect of intestinal ointment on 5-FU induced rat intestinal mucositisRats were randomly divided into normal group,model group,intestinal ointment treatment group,animal model of 5-FU induced rat chemotherapy enteroconitis,continuous administration for 8 days,daily observation of rat weight,diarrhea and food intake changes,measurement of small intestine length,Hematoxylin-eosin(HE)staining observed pathological morphological changes in small intestinal tissue in rats,while measuring the height of intestinal villi and the depth of the crypt.(3)Study on the protective mechanism of intestinal ointment on 5-FU induction of intestinal mucositis in ratsAfter the sterilization of experimental rats,the small intestinal tissue was taken,and the expression of the tight junction proteins Occludin,claudin-1 and ZO-1 of the small intestinal tissues of rats was determined by immunohistochemistry.Result:1.Compared with the control group,the gastrointestinal symptoms such as nausea,vomiting,diarrhea,TCM syndrome score,TCM syndrome total effective rate and cassette score were significantly improved in the treatment group after chemotherapy(P<0.05),which improved the quality of life of the treatment group(P<0.05).2.Intestinal ointment induces protective effect on the intestinal mucosa of rats induced by 5-FUCompared with the normal control group,the rats in the model group had reduced daily food intake,weight loss,increased diarrhea score,shortened small intestine length,and decreased villi height of duodenum,jejunum,and ileum.Loss of crypt structure and inflammatory infiltration increased,pathological score increased(P<0.05),and rats in the enteropathic treatment group increased daily feeding intake compared with the model group,weight gain,decreased diarrhea score,shortened small bowel length,improved histopathological changes,decreased pathological score(P<0.05)。3.Intestinal ointment induces protective mechanism of intestinal mucosa in rats on 5-FUCompared with the normal control group,the expression of Occludin,Claudin-1 and ZO-1 in the jejunal tissue of rats in the model group decreased(P<0.05),Compared with the model group,the expression of Occludin,Claudin-1,and ZO-1 in the jejunal tissue of rats in the enterointing treatment group increased(P<0.05)。conclusion:Intestinal ointment can improve the patient’s nausea,vomiting,diarrhea and other gastrointestinal symptoms after chemotherapy,TCM syndrome score and cassette score,the patient’s quality of life has been significantly improved,but also can improve the rat intestinal mucosal barrier damage,play a protective role,studies have shown that more with rat intestinal mucosa on the tight junction protein Occludin,Claudin-1,ZO-1 expression correlation.
