BackgroundGao Wu in the Ming Dynasty ’ acupuncture Ying ’ book said : ’ no disease and acupuncture first counter.Reverse,not even welcome.This is the first time that the word ’ inverse acupuncture ’ was recorded in the literature.The specific meaning of ’inverse moxibustion ’ refers to the pretreatment of moxibustion,which refers to the use of moxibustion drugs and heat stimulation to pre-act on human-related acupoints in the absence of disease,before the occurrence of disease or when the development of disease is still shallow,to stimulate the qi of meridians to achieve the purpose of disease prevention and health care,dredging meridians,warming meridians and dispersing cold.This method takes ’ preventive treatment of disease ’ as the main theoretical basis,and is one of the methods of disease prevention and health care of retrograde acupuncture.A large number of literature studies have proved that ’ inverse moxibustion ’ can prevent and treat diseases,strengthen physical fitness,and improve the immune function of the body.Nowadays,the incidence of malignant tumors is increasing year by year.For the treatment of such diseases,radiotherapy and chemotherapy are still the main clinical treatment,but there are still no simple and economic methods for bone marrow suppression,immune function decline and digestive system adverse reactions in clinical practice.Traditional Chinese medicine believes that ’ spleen and stomach are the root of the acquired and the vital genesis of qi and blood biochemistry ’.By intervening the acupoints related to spleen and stomach to supplement the root of the acquired,it can promote qi and blood transformation and reduce the symptoms of digestive tract discomfort.Many studies have shown that moxibustion pretreatment can significantly improve the digestive system,immune system,nervous system,reproductive system and other related diseases,and the mechanism of action may be related to moxibustion on relevant acupoints,stimulating and improving the state of meridian blood flow,regulating the content of cytokines in serum,improving the body ’s immunity,stimulating the body to produce anti-injury cytokines and other factors.Based on the ideas of “ Preventive treatment of disease ” and “ Regulating qi and blood,Treating spleen and stomach ”,combined with the pretreatment of moxibustion,this experiment explored the effects of moxibustion at different times and different acupoints on the peripheral blood image of mice,the levels of EPO,TPO and GM-CSF in serum of mice,body weight,spleen and thymus weight,and spleen and thymus index of mice.ObjectivesThe purpose of this study was to explore the effects of different intervention timings and different acupoints on the number of blood cells in peripheral blood,the levels of TPO,EPO and GM-CSF in serum,the body weight,spleen and thymus weight,spleen index and thymus index of mice were measured by the thought of“ preventive treatment of disease ” and the thought of “ regulating qi and blood,treating spleen and stomach ”,and the mechanism was discussed.so as to provide experimental basis for the intervention timing and acupoint selection of moxibustion,and provide a simple and effective treatment for the preliminary improvement of peripheral blood and immune function of patients with bone marrow suppression after chemotherapy.MethodsSixty SPF male KM mice were fed in clean laboratory for 5 days at 20 ~ 25 °C and 60 % relative humidity.Then the body weight of mice was measured,Sixty mice were randomly divided into 6 groups : blank group and model group,inverse moxibustion Piweishu group,moxibustion Zusanli group and moxibustion Piweishu group,with 10 mice in each group.After 5 days of adaptive feeding,the experiment began.The first blood collection was performed on the first day of the experiment;on the 2nd-6th day of the experiment,the Zusanli group and Piweishu group were treated with moxibustion(5 days),and the mice in each group were fixed at the same time(5 days).On the 7th day of the experiment,the second blood was collected.On the 8 th-10 th day(3 days)of the experiment,the mice in the model group,the inverse moxibustion Zusanli group,the inverse moxibustion Piweishu group,the moxibustion Zusanli group and the moxibustion Piweishu group were given cyclophosphamide(CTX)intraperitoneal injection,and the mice in the blank group were given the same dose of normal saline.The third blood collection was performed on the 11 th day of the experiment;the mice in Zusanli group and Piweishu group were treated with moxibustion(5 days)from the 12 th to 16 th day of the experiment.The mice in each group were grabbed and fixed at the same time(5 days).The mice were weighed on the 17 th day of the experiment.The fourth blood collection was carried out and the mice were sacrificed.The spleen and thymus were removed.The number of blood cells in peripheral blood of mice in each group was detected by automatic blood cell analyzer.The levels of TPO,EPO,granulocyte-phagocyte colony stimulating factor and body weight were detected by ELISA.spleen and thymus index were measured.Results1.Determination of white blood cell(WBC),red blood cell(RBC),hemoglobin(HGB),platelet(PLT)by automatic blood cell analyzer :(1)WBC :After CTX modeling,except the blank group,WBC decreased remarkably,suggesting that bone marrow suppression modeling was successful.On the 17 th day of the experiment,the WBC in peripheral blood of model group,reverse moxibustion group and moxibustion group showed an upward trend.The upward trend of reverse moxibustion group was higher than that of model group,and the reverse moxibustion group was more obvious.The rising trend of moxibustion group was lower than that of model group.Compared with the mode group,there was no statistical difference in WBC before and after moxibustion in the inverse moxibustion group and moxibustion group(P > 0.05).The change of WBC before and after moxibustion was statistically significant(P < 0.05).(2)RBC : On the 17 th day of the experiment,the dynamic trend analysis of RBC showed that the moxibustion Zusanli group and the moxibustion Piweishu group had a significant effect on slowing down the decline of RBC compared with the reverse moxibustion Zusanli group;compared with the reverse moxibustion Zusanli group,moxibustion Zusanli group,moxibustion Piweishu group RBC before and after moxibustion changes were statistically significant(P < 0.05);compared with the model group,the changes of HGB before and after moxibustion were very significant(P < 0.05),but the downward trend of HGB in the spleen-stomach-shu group was almost coincident with the model group,and the reverse moxibustion group was significantly lower than the model group.(4)PLT : On the 17 th day of the experiment,except the blank group,PLT of mice in each group showed an upward trend;compared with model group,the PLT of moxibustion group and reverse moxibustion group elevated significantly,but there was no difference in PLT before and after reverse moxibustion(P > 0.05).Compared with the inverse moxibustion group,the changes of PLT before and after inverse moxibustion were significant(P < 0.05).2.The levels of erythropoietin(EPO),thrombopoietin(TPO)and granulocyte colony-stimulating factor(GM-CSF)in serum were determined by enzyme-linked immunosorbent assay(ELISA):(1)EPO : On the 17 th day of the experiment,compared with the blank group,the EPO levels of the model group,the inverse moxibustion group and the moxibustion group decreased(P < 0.05),and the EPO level of the inverse moxibustion group was higher than that of the moxibustion group(P < 0.05).The EPO level of the spleen-stomach point selected by the inverse moxibustion group and the moxibustion group was higher than that of the Zusanli point(P < 0.05).(2)TPO : On the 17 th day of the experiment,compared with the blank group,the TPO levels in the model group,inverse moxibustion group and moxibustion group decreased(P < 0.05);compared with the model group,the level of TPO in the moxibustion group was significantly decreased(P < 0.05).(3)GM-CSF : On the 17 th day of the experiment,compared with the model group,the GM-CSF level in the inverse moxibustion group was decreased(P < 0.05),and the GM-CSF level in the moxibustion group was increased(P < 0.05).Compared with the moxibustion group,the GM-CSF level in the moxibustion group was decreased(P < 0.05).3.(1)Body weight : Comparing with the model group,there was no significant difference in body weight between the moxibustion group and the model group(P >0.05).The body weight of the reverse moxibustion group was slightly lower than that of the moxibustion group,and the weight and index of the spleen and thymus of the mice were analyzed.(2)Spleen weight : Comparing with the model mice,the spleen weight of the inverse moxibustion group was increased(P < 0.05);(3)Thymus weight : Compared with the model rats,the thymus weight the reverse moxibustion group and the moxibustion group increased(P < 0.05);(4)Spleen index :Compared with the model group mice,the spleen index of the reverse moxibustion group was increased,and the reverse moxibustion group was statistically significant(P < 0.05).with the anti-moxibustion Zusanli group,the spleen indexes of mice in the anti-moxibustion Piweishu group and the moxibustion group decreased(P <0.05).(5)Thymus index number : Compared with model group,thymus index of reverse moxibustion group and moxibustion group increased(P < 0.05);compared with the anti-moxibustion Zusanli group,moxibustion Zusanli group and moxibustion Piweishu group,the thymus index number of mice in the anti-moxibustion Piweishu group was more higher(P < 0.05).ConclusionCompared with the moxibustion Piweishu group,the effect of reverse moxibustion Zusanli group on promoting the rise of WBC was significant.Compared with the moxibustion group,the effect of reverse moxibustion Zusanli group on promoting the rise of PLT was significant.Compared with reverse moxibustion zusanli group,moxibustion group had a significant effect on decreasing RBC and HGB.The TPO level of moxibustion at Piweishu was significantly lower than that of model group.The EPO level and GM-CSF level of moxibustion at Piweishu were the highest,and the spleen index of moxibustion at Zusanli was the highest,and the thymus index of moxibustion at Piweishu was the highest.This conclusion shows that moxibustion pretreatment can regulate qi and blood by affecting the number of peripheral blood cells and serum cytokines,improve immune function through the spleen and thymus,reduce bone marrow suppression reaction,in line with the theory of ’ preventive treatment of disease ’ and ’ spleen and stomach,regulating qi and blood’ in traditional Chinese medicine.
