Background and purposeColorectal cancer is one of the most common malignant tumors.According to the data provided by the international agency for cancer research,colorectal cancer is the third most common malignant tumor,and mortality ranks second among cancer-related deaths.SPOCD1 is a newly discovered gene encoding a protein belonging to the transcription factor S-Ⅱ(TFIIS)family.Studies have shown that the expression of the SPOCD1 is closely related to tumor proliferation,invasion,and migration.However,the prognostic value and potential function of the SPOCD1 in colorectal cancer have not been studiede.The purpose of this study is to further explore the clinical value of SPOCD1 in colorectal cancer and its clinical informaticsMethodsFirstly,we obtained the transcriptome data of colorectal cancer from the TCGA database,GTEx database,and GEO database,analyzed the expression difference of SPOCD 1 between colorectal cancer tissue and normal tissue,and verified the expression of SPOCD1 in several common colorectal cancer cell lines by RT-qPCR.Additionally,we obtained the prognosis information of colorectal cancer patients from the TCGA database and analyzed the correlation between SPOCD 1 expression and the prognosis of colorectal cancer patients.Then,we analyzed the methylation level of SPOCD1 in colorectal cancer through the cBioPortal platform;Methsurv was used to explore the relationship between the methylation level of SPOCD1 and the prognosis of colorectal cancer patients.Finally,we explored the cellular components,biological functions,molecular functions,and signal pathways involved in the SPOCD1 by Gene Set Enrichment Analysis(GO analysis and GSEA analysis),and analyzed the relationship between SPOCD1 expression and immune infiltration in colorectal cancer by TIMER 2.0 platform.ResultsCompared with normal tissues,the SPOCD1 was highly expressed in colorectal cancer tissues(P<0.001).RT-qPCR results also showed that SPOCD1 was highly expressed in common colorectal cancer cell lines,especially in MSI colorectal cancer cell lines(P<0.05).Kaplan-Meier survival analysis showed that high expression of SPOCD1 was associated with a poor prognosis of colorectal cancer(HR=1.61,95%confidence interval:1.14-2.28,P<0.05).The methylation level of SPOCD1 in rectal cancer was lower than that in normal tissues(p<0.05),and the overall survival of patients with hypomethylated SPOCD1 was shorter than that of patients with hypermethylated colorectal cancer(p<0.05).GSEA enrichment analysis showed that SPOCD1 expression was related to apical-junction pathway,epithelial-mesenchymal transition pathway(EMT),inflammatory response pathway,interferon-gamma response pathway,and KRAS signaling pathway.By analyzing the immune cell infiltration level related to the SPOCD1,it shows that the change in SPOCD1 expression is related to the changes of various types of immune cells in colorectal cancer,which has a significant impact on the tumor microenvironment of colorectal cancer.ConclusionThis study confirmed that the overexpression of the SPOCD1 is closely related to the poor prognosis of colorectal cancer.SPOCD1 is expected to become a new biological research target a new therapeutic target,and a potential prognostic biomarker of colorectal cancer.
