| Mica is the most common silicate mineral.The fermentation extract of organic mica is obtained from Huanggang rock and green plum under appropriate temperature conditions.It has antibacterial,anti-inflammatory,antioxidant and anti-tumor effects.Inflammation,as the basic pathological process of many diseases,has the characteristics of rapid onset and inducing chronic diseases.When it is serious,it will cause irreversible damage to the human body.The National Cancer Research Center has reported that 1/6 of all cancers in the world are caused by inflammation caused by bacteria and viruses.Infection with hepatitis B or hepatitis C virus will cause patients to suffer from hepatitis,causing long-term repeated damage and repair of the liver and increasing the risk of liver cancer.Liver cancer is one of the most common malignant tumors,and more and more patients suffer from the pain of liver cancer.Therefore,the treatment of liver cancer has become the focus of medical attention.The abnormal activation of phosphatidylinositol 3-kinase/protein kinase B/mammalian target protein of rapamycin(PI3K/Akt/m TOR)signaling pathway is closely related to the occurrence of hepatocellular carcinoma,providing a potential target for targeted therapy.The aim of this study was to investigate the anti-inflammatory effect of organic mica fermentation extract and the effect of PI3K/Akt/m TOR signal pathway on inducing apoptosis and proliferation and migration of hepatoma cells.Firstly,the anti-inflammatory activity of the organic mica fermentation extract was verified,and the MTT test verified the effect of the organic mica fermentation extract on the activity of RAW264.7 cells.Then,from the perspective of inflammation theory,the mouse monocyte macrophage cell line(RAW264.7)was induced by lipopolysaccharides(LPS),Subsequently,whether the organic mica fermentation extract has anti-inflammatory effect was judged by the detection of the release of nitric oxide(NO)and the detection of pro-inflammatory cytokines.Subsequently,the inhibitory effect of organic mica fermentation extract on human hepatoma cell line(Hep G2)was verified.The effect of organic mica fermentation extract on the activity of Hep G2 cells was detected by culturing Hep G2 cells in vitro;At the same time,the molecular mechanism of the influence of organic mica fermentation extract on hepatoma cells was determined by measuring the changes of signal molecule expression and activity in cells before and after the application of organic mica fermentation extract.After being treated with different concentrations of organic mica fermentation extract,MTT assay was used to verify the effect of organic mica fermentation extract on the activity of Hep G2 cells and normal human liver cell line(L02);Cell migration was detected by cell scratch test;Then TUNEL cell apoptosis assay was used to detect the early apoptosis of HCC cells;Subsequently,the expression of PI3K/Akt/m TOR signaling pathway related genes and apoptosis gene BCL2 were detected by QRT PCR.Finally,the expression levels of m TOR,Akt and PI3 k proteins were detected by Western blot.Results: in the anti-inflammatory experiments,the organic mica fermentation extract had no obvious toxic effect on raw264.7 cells at the concentrations of 1% and 2%.The organic mica fermentation extract can reduce the level of nitric oxide secreted by raw 264.7 cells stimulated by LPS,and can reduce the inflammation related tumor necrosis factor in raw 264.7 cells,TNF-α(Tumor necrosis factor α)、 IL-1α(Interleukin-1α)、IL-1β(Interleukin-1β)、The m RNA expression level of IL-6(Interleukin-6).In the anti-tumor related experiments,the organic mica fermentation extract has no obvious toxic effect on L02 cells at the concentration of 1% and 2%,and the toxicity to Hep G2 cells is greater than that to L02 cells.Compared with the control group,the proliferation and migration ability of Hep G2 cells in the 1% and 2% organic mica fermentation extract solution treatment groups were significantly reduced.After 48 hours of treatment,Hep G2 cells could significantly induce apoptosis of hepatoma cells,and could reduce the m RNA expression levels of PI3K/Akt/m TOR related pathway genes m TOR,Akt,PI3 K and apoptosis gene BCL2 in Hep G2 cells.Western blot results showed that the expression levels of m TOR,Akt and PI3 K proteins were decreased,The downregulation of protein level suggested that the inhibitory effect of organic mica fermentation extract on Hep G2 cells might be caused by regulating PI3K/Akt/m TOR signaling pathway.In conclusion,the organic mica fermentation extract can reduce the cellular inflammation induced by lipopolysaccharide;And can inhibit the proliferation of hepatoma cells,showing a significant inhibitory effect on hepatoma,and to a certain extent,it is related to PI3K/Akt/mTOR pathway. |
