| Objective: After dopamine cells induced by 6-Hydroxydopamine(6-OHDA)were damaged,the effects of Icaritin(ICT)on apoptosis-related proteins Bax/Bcl-2,Caspase-9,Cleaved Caspase-3were observed,and the protective effect of ICT on dopaminergic neurons was explored,to provide theoretical basis for ICT treatment of Parkinson’s disease(PD).Methods: 1)We divided randomly into Control group,ICT 0.01 μM group,6-OHDA group,6-OHDA+ ICT 0.001 μM group and 6-OHDA+ ICT 0.01 μM group.2)After ICT pre-processing for 1 hour,add 6-OHDA for 24 hours.3)The apoptosis rate was detected by flow cytometry.4)The expressions of α-Syn,TH,Bax/Bcl-2,Caspase-9,Cleaved Caspase-3,in 6-OHDA-induced dopaminergic neurons were detected by Western blot.Results: Compared with the blank group,the apoptosis rate detected by flow cytometry in the 6-OHDA group was significantly increased.and the expression of antiapoptotic protein Bcl-2 decreased.After pretreatment with ICT for 1 h,the apoptosis rate decreased,and the protein levels of α-Syn and pro-apoptotic proteins Bax/Bcl-2,Caspase-9 and Cleaved Caspase-3 decreased significantly.The amount of TH increased.Conclusion: 6-OHDA-induced dopaminergic neuron can be protected from damaging by inhibiting apoptosis through ICT,which is relevant to the expression of apoptosis related protein Bax/Bcl-2,Caspase-9 and Cleaved Caspase-3. |
