| Objective:Cervical cancer is a common gynecological tumor.Patients with advanced or recurrent cervical cancer need platinum based standard chemotherapy.However,in the process of chemotherapy,some patients produce cisplatin tolerance through endogenous or acquired mechanisms,which seriously restricts the chemotherapeutic effect of cisplatin on cervical cancer and affects the prognosis of cervical cancer patients.Therefore,it is very important to study the potential mechanism of drug resistance of cervical cancer and improve the effect of chemotherapy.Previous studies have found that long-chain noncodingRNA H19 is highly expressed in most tumor drug-resistant tissues or cells,but the relationship between long-chain noncodingRNA H19 and drug resistance of cervical cancer is inconclusive.Therefore,this study aims to explore the role of long-chain noncodingRNA H19 in the process of drug resistance of cervical cancer and preliminarily explore the possible mechanism of long-chain noncodingRNA H19 in regulating drug resistance of cervical cancer.Method:1.He La cisplatin resistant cell lines He La / DDP and He La cell lines were cultured,and the expression level of lncrna H19(nr_002196.2)was detected by q PCR.2.Lncrna H19 interference vector was constructed,packaged into lentivirus and infected with He La cisplatin resistant cell line He La / DDP.The infection efficiency was verified by q PCR,and He La cisplatin resistant cells interfering with lncrna H19 were stably screened.3.Western blot was used to detect the expression level of Nrf2 in cells of blank control group(Hela / DDP contral),empty group(Hela/ DDP NC)and interference group(Hela / DDP siRNA).4.According to the grouping,the cells were treated with cisplatin(DDP),and the cell proliferation was detected by CCK-8 method after 24 hours;result:1.QPCR results showed that the expression level of lncrna H19 in He La /DDP cell group was significantly higher than that in He La cell group(P < 0.05)2.Through siRNA interference experiment,the expression of lncrna H19 in He La / DDP cells decreased,and the expression level of Nrf2 also decreased significantly by Western blot experiment,indicating that lncrna H19 may regulate the expression of Nrf2.3.The proliferation rate of helnck cells was increased by the addition of cisplatin,and it was found that the proliferation rate of helnck cells was decreased by the addition of cisplatin.Conclusion: lncrna H19 is related to the drug resistance of cisplatin resistant cervical cancer cells.Knocking down the expression of long-chain noncodingRNA H19 can improve the sensitivity of He La / DDP cells to cisplatin.Long-chain noncodingRNA H19 may lead to the drug resistance of cervical cancer by regulating the expression of Nrf2. |
