| Objective:This paper investigate the expression and significance of toll like receptor 4(TLR-4)and human β defensin 2(HBD-2)fetal membrane tissue in pregnant women with premature rupture of membranes(PROM)in Qinghai area,and provide some experimental basis for PROM control in this area.Method:(1)We collected 30 cases of fetal membrane tissue from pregnant women with premature rupture of membranes(PROM group)and 30 cases from normal pregnant women(normal group)who delivered at the same time in Qinghai University Affiliated Hospital from November 2020 to November 2021.There were no other obstetric-related comorbidities in the two groups of pregnant women;(2)The localization and expression of TLR-4 and HBD-2 in fetal membrane tissue were detected by immunohistochemical staining,and the expression of TLR-4 and HBD-2in fetal membrane tissue were detected by Western blot;(3)The independent samples t test was used to analyze whether there were differences in the expression of TLR-4and HBD-2 in fetal membranes tissue between the two groups,and Pearson correlation was used to analyze the correlation between the two factors.Results:(1)Immunohistochemical staining showed that the localization of TLR-4and HBD-2 in fetal membrane tissue was basically the same in the two groups,and they were located in the cytoplasm of amniotic epithelial cells and chorionic trophoblast cells.The average optical density of TLR-4 and HBD-2 in the PROM group was higher than that in the normal group,and there was a statistical difference(P < 0.05);(2)Western blotting experiments showed that the expression levels of TLR-4 and HBD-2 in the PROM group were higher than those in the normal group,with statistical differences(P < 0.05);(3)Western blot findings that there was a positive correlation between TLR-4 and HBD-2 protein expression in PROM group(r=0.380,P=0.039,P<0.05),but there was no significant correlation between the expression of TLR-4 and HBD-2 in fetal membranes of normal pregnancy(r=0.033,P=0.863,P>0.05).Conclusion:(1)The localization of TLR-4 in PROM group and normal pregnancy group in Qinghai was basically the same.The expression level of TLR-4 protein in PROM group was higher than that in normal pregnancy group;(2)The localization of HBD-2 in PROM group and normal pregnancy group in Qinghai was basically the same.The expression level of HBD-2 protein in PROM group was higher than that in normal pregnancy group;(3)The protein expression levels of TLR-4 and HBD-2 were positively correlated in the PROM group in Qinghai,but there was no significant correlation between the two factors in the normal pregnancy group,and considered that TLR-4 and HBD-2 may be related to the pathophysiology of PROM membrane rupture in this region. |
