Objectives: To research the impact and mechine-made of miR-95-5p on the osteogenic differentiation of adipose stem cells(ADSCs)by targeting bone morphogenetic protein2(BMP2).Methods: ADSCs were cultured and divided into the miR-NC group(transfected with NC mimic)and miR-95-5p group(transfected with miR-95-5p mimic).The ALP activity and staining was detected by kit and microscope,respectively.And the level of A570 was detected by alizarinred staining.The expression levels of BMP2,Runx2 and osteocalc in(OCN)were detected by Westernblotting.The miR-95-5p targeting BMP2 gene was verified by luciferase reporter gene experiment.Results: Compared with the miR-NC group,the ALP activity,ALP staining,A570 nm level of alizarinred staining,BMP2,Runx2,OCN expressionin the miR-95-5p group were significantly decreased after osteogenic differentiation(P<0.05).miR-95-5p targeted the 868 to 874 base of BMP2 gene m RNA3’UTR.Compared with the miR-NC group,the fluorescence activity of wild-type BMP2 luciferase reporter gene in ADSCs of miR-95-5p group was significantly decreased(P<0.05).Conclution:miR-95-5p inhibits the osteogenic differentiation of ADSCs,and targeting BMP2 gene is a possible molecular mechanism.
