Objective: through high-throughput sequencing technology and bioinformatics method,this study intends to find lncrnas differentially expressed in samples and related to renal cell carcinoma.Through the prediction and functional analysis of target genes of target lncrnas,this study explores the possible biological functions of target lncrnas,and strives to further improve the regulatory network of lncrnas in the occurrence and development of renal cell carcinoma,It provides a theoretical basis for the search for specific tumor markers of renal cell carcinoma and effective drug treatment targets.Methods: ten human cancer tissues(C1-C10)and adjacent tissues(N1-N10)were sequenced by Illumina sequencing platform.Through intra group and inter group comparison,taking the expression of lncrna fpkm as the reference standard,the overall lncrna with significant difference between cancer tissues and adjacent tissues was obtained by correlation statistical analysis.MNDRV3.1 gene platform to screen lncrnas with significant expression differences related to renal cancer,further predict the target genes of lncrnas with significant expression differences related to renal cancer according to the positional relationship between lncrna and m RNA,and analyze the function enrichment of target genes.By classifying the functions of target genes,we can understand the specific role of differential lncrna in the occurrence and development of renal cell carcinoma.Results:1.In this experiment,10 human renal cell carcinoma and adjacent tissue samples were sequenced and analyzed by high-throughput sequencing technology.Through the comparative analysis between cancer tissues and adjacent tissues,20 significantly differentially expressed lncrnas related to renal cell carcinoma were obtained.Among them,lncrna10 were significantly up-regulated and lncrna10 were significantly down regulated in renal cell carcinoma.2.The 10 significantly up-regulated lncrnas obtained in this study were PVT1,LINC00887,FTX,LUCAT1,LINC-PINT,LINC01094,FER1L4,DGCR5,CDKN2B-AS1,HIF1A-AS2.The following results were obtained by target gene prediction of 10 lncrnas.(1)PVT1: one corresponding target gene: MYC.(2)LINC00887: four corresponding target genes: ATP13A3,CPN2,GP5 and LRRC15.(3)FTX: one corresponding target gene: ZCCHC13.The target gene has no obvious function and pathway enrichment.(4)LUCAT1: two corresponding target genes: ARRDC3 and ADGRV1.(5)LINC-PINT: a corresponding target gene: MKLN1.(6)LINC01094: three corresponding target genes: ANXA3,BMP2 K and FRAS1.(7)FER1L4: three corresponding target genes: CPNE1,NFS1 and RBM12.Among them,the target gene RBM12 was not enriched in GO function and KEGG pathway.(8)DGCR5: 4 corresponding target genes: PRODH,DGCR6,DGCR2,AC0073264。 Target gene AC0073264 no GO function and KEGG pathway enrichment were found.(9)CDKN2B-AS1: one corresponding target gene: CDKN2 A.(10)HIF1A-AS2: 3 corresponding target genes: AL3559163、SNAPC1、HIF1A。Target gene AL3559163 did not find GO function and KEGG pathway enrichment.3.The 10 significantly down regulated lncrnas obtained in this study were NPSR1-AS1,LINC02121,TARID,DLEU1,AC024022.1,COL18A1-AS1,HULC,TCL6,DRAIC,WT1-A.The following results were obtained by target gene prediction of 10 lncrnas.(1)NPSR1-AS1: two corresponding target genes: NPSR1 and DPY19L1.(2)LINC02121: one corresponding target gene: ADAMTS16.(3)TARID: one corresponding target gene: TBPL1.(4)DLEU1: two corresponding target genes: DLEU7 and RNASEH2 B.No enrichment of go function and KEGG pathway was found in the target gene DLEU7.(5)AC024022.1: There are 2 corresponding target genes: SHISA3 and ATP8A1.No enrichment of GO function and KEGG pathway was found in the target gene SHISA3.(6)COL18A1-AS1: two corresponding target genes: SLC19A1 and COL18A1.(7)HULC: one corresponding target gene: SLC35B3.(8)TCL6: two corresponding target genes: TCL1 B and TCL1 A.No enrichment of GO and KEGG was found in target gene TCL1 B.(9)DRAIC: 2 corresponding target genes: AC0272371,RPLP1.Target gene AC0272371.no functional enrichment of GO and KEGG was found.(10)WT1-A: one corresponding target gene: WT1.Conclusion:1.In this experiment,10 human renal cell carcinoma and adjacent tissue samples were sequenced and analyzed by high-throughput sequencing technology.Through the comparative analysis between cancer tissues and adjacent tissues,20 significantly differentially expressed lncrnas related to renal cell carcinoma were obtained,the target genes corresponding to 20 lncrnas were found,and the function and pathway of the target genes were enriched.2.10 significantly up-regulated lncrnas associated with renal cell carcinoma were PVT1,LINC00887,FTX,LUCAT1,LINC-PINT,LINC01094,FER1L4,DGCR5,CDKN2B-AS1,HIF1A-AS2.(1)LINC00887-ATP13A3,LUCAT1-ARRDC3,FER1L4-CPNE1,DGCR5-PRODH,LUCAT1-ARRDC3,HIF1A-AS2--HIF1 A participate in many processes such as material exchange and energy metabolism;(2)PVT1-MYC,LINC01094-ANXA3,LINC01094-BMP2 K,PVT1-MYC,LINC-PINT-MKLN1,HIF1A-AS2--HIF1 A are involved in the development of kidney,collecting system and other tissues;(3)PVT1-MYC participates in Erb B signaling pathway and MAPK signaling pathway,CDKN2B-AS1--CDKN2 A participates in p53 signaling pathway,HIF1A-AS2--HIF1 A participates in m TOR signaling pathway,which are closely related to the occurrence and development of renal cell carcinoma,and may be the focus of renal cell carcinoma research in the future.It may be an important breakthrough in finding therapeutic targets for renal cell carcinoma.3.10 lncrnas significantly down regulated in renal cell carcinoma were NPSR1-AS1,LINC02121,TARID,DLEU1,AC024022.1,COL18A1-AS1,HULC,TCL6,DRAIC,WT1-AS.(1)TARID-TBPL1,NPSR1-AS1--NPSR1,DLEU1-RNASEH2 B,AC024022.1-ATP8A1,COL18A1-AS1--SLC19A1,COL18A1-AS1--COL18A1,HULC-SLC35B3participate in multiple material exchange and energy metabolism processes;(2)LINC02121-ADAMTS16,TCL6-TCL1 B,WT1-AS--WT1 are involved in the development of kidney and collecting system and other tissues;(3)DLEU1-RNASEH2 B,DRAIC-RPLP1,HULC-SLC35B3 may be used as predictors of good prognosis of renal cell carcinoma.
