| Objective: Curcumin was used to intervene the diabetic periodontitis rats.The body weight,fasting blood glucose,periodontal tissue examination,alveolar bone microstructure and inflammatory factors in Micro-CT were analyzed.The control effect of curcumin on diabetic periodontitis rats and the protective effect of periodontal tissue were evaluated.Methods:a total of 45 male SD rats were randomly selected from 7 randomly selected groups.The rats were fed with normal diet as the A group(blank control group).The remaining 38 rats were fed with high sugar and high-fat diet for +STZ abdominal cavity injection.21 rats after successful modeling were randomly divided into three groups: group B(model group n = 7): 0.5% sodium carboxymethylcellulose(2ml / kg / day);Group C(curcumin group n = 7): Curcumin suspension(200mg / kg / day),group D(dimethylbis group n = 7): metformin(200mg / kg / day).According to the above standards,the experiment was intervened for 6W,and the blood glucose and body weight were monitored for 1W / time;After 6 weeks,the rats were killed,and the clinical related indexes of periodontal tissue were measured: gingival index(GI),mobility(TM),periodontal pocket probing depth(PPD).IL-6 and TNF in rat serum were detected by ELISA-α And IL-1 β、PGE2 levels of various inflammatory factors,CT,trap and other factors;Micro CT scanning was used to analyze the absorption and microstructure changes of alveolar bone in rats.Result:(1)Blood glucose: after 6 weeks of drug intervention,there was significant difference among the four groups(P < 0.05).There was no significant difference between C and D(P > 0.05),and there were significant differences between the other two groups(P< 0.05).A.There was no significant difference in blood glucose between 6 weeks after treatment and before treatment in group B(P > 0.05),and blood glucose decreased significantly in groups C and D(P < 0.05).(2)Clinical examination of periodontal tissue:GI and PPD: after 6 weeks of drug intervention,there was significant difference among the four groups(P < 0.05).There was no significant difference between C and D(P > 0.05),and there were significant differences between the other two groups(P < 0.05).A.There was no significant change in GI value in group B before and 6 weeks after treatment(P >0.05).GI index and PPD decreased significantly in groups C and d after 6 weeks of treatment(P < 0.05).Tm: there was no significant difference between groups A and B(P <0.05).There was no significant difference between the four groups before and after treatment(P > 0.05).(3)Micro CT: the alveolar bone mass ratio(BV / TV),trabecular thickness(TB.Th)and bone mineral density(BMD)of maxillary first molar in groups C and D were significantly higher than those in group B(P < 0.05),and the Trabecular separation(TB.SP)was significantly lower than that in group B(P < 0.05),but the number of trabeculae(TB.N)was not significantly different from that in group B(P > 0.05);The distance between enamel cementum boundary and alveolar ridge top(acj-ac)was significantly lower than that in group B(P < 0.05).There was no significant difference between groups C and D(P > 0.05).(4)ELISA: the concentrations of IL-1B,IL-6,PGE2,TNF-a and trap in serum of rats in groups C and D were significantly lower than those in group B model group(P < 0.05),and the concentration of CT was increased(P < 0.05);The decrease of TNF-a and trap in group D was more significant than that in group C(P <0.05).Conclusion:(1)Successful establishment of diabetic periodontitis rat model.(2)Curcumin can reduce the fasting blood glucose of diabetic periodontitis rats to some extent.(3)Curcumin can reduce periodontal inflammation(GI and PPD)in diabetic periodontitis rats,but has no effect on tooth mobility(TM).(4)Curcumin can improve the alveolar bone microstructure of diabetic periodontitis rats.(5)Curcumin can reduce the secretion of IL-1B,IL-6,PGE2,TNF-A and TRAP,and promote the secretion of CT in diabetic periodontitis rats. |
