Objective:The purpose of the experiment is to explore the distribution of 30 erythrocyte antigen alleles in 12 rare blood group systems in Mongolian and Han people in Inner Mongolia RH(C/c/E/e)、Lw(Lw~a/Lw~b)、Duffy(Fy~a/Fy~b)、Kidd(Jk~a/Jk~b)、MNS(M/N/S/s/Mur)、Scianna(Sc2)、Colton(Co~a/Co~b)、Dombrock(Do~a/Do~b)、Kell(K/k/Kp~a/Kp~b)、Diego(Di~a/Di~b)、Yt(Yt~a/Yt~b)and Lutheran(Lu~a/Lu~b),for the study of hematology of Mongolian and Han people,precise blood transfusion,clinical science and rational use of blood and providing data support to the construction of the rare blood group bank.Methods:(1)Through the HIS system,we searched 220 Mongolian inpatients and 198 Han inpatients in our hospital.(2)Use the nucleic acid purifier to extract DNA samples from 220Mongolian and 198 Han people.(3)Use the UV spectrophotometer to determine the purified extracted DNA samples of Mongolian and Han people in Inner Mongolia area,and the DNA concentration range(40-80)ng/μL,the OD260/OD280 is 1.6-2.0.(4)The fluorescence PCR was used to detect the 30 red blood cell antigen alleles of the 12 red blood cell blood group systems of 220 Mongolian and 198 Han people in Inner Mongolia whose DNA concentration meets the experimental requirements,and the results of the detection were statistically analyzed using SPSS 23.0 statistical software.Results:(1)The gene frequencies of the 12 rare blood group in the Mongolian population in Inner Mongolia are as follows.Among them,the gene frequency of the RHCE blood group system is:C=0.6136,c=0.3864,E=0.2659,e=0.7341;The frequency of MNS blood group gene is:M=0.6091,N=0.3909,S=0.0636,s=0.9318,Mur=0;the Duffy blood group gene frequency is:Fy~a=0.8568,Fy~b=0.1432;the Kidd blood group gene frequencyis:Jk~a=0.5227,Jk~b=0.4773; the Diego blood group gene frequency is:Di~a=0.0273,Di~b=0.9727;the Dombrock blood type gene frequency is:Do~a=0.1636,Do~b=0.8364;the Kell blood group gene frequencyis:K=0.0023,k=0.9977,kp~a=0.0091,Kp~b=0.9909;the frequency of Yt blood group genes is:Yt~a=0.9864,Yt~b=0.0136;the frequency of Lw blood group gene is:Lw~a=1,Lw~b=0;the frequency of Scianna blood group gene is:Sc2=0;the frequency of Colton blood group gene is:Co~a=1,Co~b=0;the frequency of Lutheran blood group gene is:Lu~a=0,Lu~b=1.(2)The frequency of the 12 rare blood group genes in the Han population in Inner Mongolia is as follows:the gene frequency of the RHCE blood group system is:C=0.6060,c=0.3940,E=0.3031,e=0.6969;the gene frequency of the MNS blood group system is:M=0.4470,N=0.5530,S=0.0455,s=0.9545,Mur=0.0101;the gene frequency of the Duffy blood group system is:Fy~a=0.9318,Fy~b=0.0682;the gene frequency of the Kidd blood group system is:Jk~a=0.5152,Jk~b=0.4848;the gene frequency of the Diego blood group system is:Di~a=0.0303,Di~b=0.9697;the gene frequency of the Dombrock blood group system is:Do~a=0.1288,Do~b=0.8712;the gene frequency of the Lw blood group system is:Lw~a=1,Lw~b=0;the gene frequency of the Scianna blood group system is:Sc2=0;the gene frequency of the Kell blood group systemis:K=0,k=1,kp~a=0,Kp~b=1;the gene frequency of the Colton blood group system is:Co~a=1,Co~b=0;the gene frequency of the Lutheran blood group system is:Lu~a=0,Lu~b=1;the gene frequency of the Yt blood group system is:Yt~a=0.9975,Yt~b=0.0025.(3).After calculation,we found that in the MNS blood group system of the Mongolian population in this region,and the frequency of the M allele is higher than that of the Han in this region.The frequency of N allele was lower than that of Han in Inner Mongolia,the difference was statistically significant(p<0.01);in the Duffy blood group system of Mongolian population in this region,the frequency of Fy~aallele was higher than that of Han in this region.The frequency of Fy~ballele was lower than that of Han ethnic group in this area,and the difference was statistically significant(p<0.05).Conclusion:(1)The RHCE,MNS,Duffy,Kidd and Dombrock blood types of Mongolian popul-ation in Inner Mongolia are polymorphic and have their unique distribution characteristics.(2)The RHCE,MNS,Duffy,Kidd and Dombrock blood types of the Han population in this area are polymorphic and have their unique distribution characteristics.(3)The blood group systems-Kell,Yt,Diego,Lw,Scianna,Colton,and Lutheran of Mongolian and Han people in Inner Mongolia are monomorphic,which are similar to the distribution of people in other parts of our country.(4)Improve the clinical awareness of rare blood types,reduce the occurrence of ineffective blood transfusion in patients who have produced red blood cell alloantibodies,and truly achieve accurate blood transfusion.
