Effects Of Xiaoyao Pill Combined With Guipi Decoction On Behavior,Hippocampal 5-HT,DA,BDNF And Intestinal Flora In Rats With Postpartum Depression

ObjectiveBy detecting the effects of Xiaoyao Pill combined with Guipi Decoction on the behavior,5-HT,DA,BDNF expression and intestinal flora structure of postpartum depression(PPD)rats,and to explore its efficacy and possible mechanism.MethodsFifty-six qualified SD female rats were screened by open field test and divided into blank group,sham operation group and PPD model group according to the random number table method.The rats in the PPD model group underwent bilateral ovariectomy,and those who were qualified after the operation were injected with hormones continuously,and the hormones were suddenly stopped after 24 days to prepare the PPD model.After modeling,blank group,sham operation group,and PPD model group were subjected to sucrose preference test,open field test,and forced swimming test to evaluate the model.The PPD model group was randomly divided into model group,fluoxetine group,Xiaoyao Pill combined with Guipi Decoction low-dose,medium-dose and high-dose groups,respectively given corresponding drugs for 3-week intragastric intervention,blank,sham operation and model groups given an equal volume of distilled water by gavage.Then,the body weight level and behavioral changes of the rats were recorded,the expressions of 5-HT,DA and BDNF in the hippocampus of the rats were detected by ELISA,and the structural changes of the intestinal flora of the rats were detected by high-throughput sequencing technology.Results1 Body weight of rats: Compared with the model group,the body weight of the rats in the low,medium and high-dose groups of Xiaoyao Pill combined with Guipi Decoction increased(P<0.01).2 Behavioral results: Compared with the model group,the sucrose preference rate,number of crossing and rearing of the rats in Xiaoyao Pill combined with Guipi Decoction low,medium and high-dose groups increased(P<0.05,P<0.01),swimming immobility time Compared with the fluoxetine group,the number of rearing of the rats in Xiaoyao Pill combined with Guipi Decoction high-dose group increased(P<0.05).3 ELISA test results: Compared with the model group,the expressions of 5-HT,DA and BDNF in the hippocampus of the rats in Xiaoyao Pill combined with Guipi Decoction low,medium and high-dose groups increased(P<0.01).Compared with the fluoxetine group,the expression of DA in the Xiaoyao pill combined with the high-dose Guipi decoction group increased(P<0.01).4 High-throughput sequencing detection results:(1)α diversity: Shannon,Simpson,ACE,Chao 1 index results indicated that the α diversity of the intestinal flora of rats in the model group did not change significantly(P>0.05);The low,medium and high-dose groups of Combined Guipi Decoction also had no significant effect on the α diversity of model rats(P>0.05).(2)β diversity: PCo A,PCA,UPGMA,and MRPP analysis showed that there were obvious differences in the community composition between the blank group and the model group;There were differences in the community composition of rats between the two groups,which were significantly different from those in the low and high-dose groups(P<0.05).(3)Species composition analysis: At the phylum level,the relative abundance of Bacteroidota in the model group decreased(P<0.05),while the relative abundances of Desulfobacterota and Chloroflexi increased(P<0.01);compared with the model group,the relative abundance of Bacteroidota decreased(P<0.05),the relative abundance of Bacteroidota increased in the low and high-dose groups of Xiaoyao Pills combined with Guipi Decoction(P<0.05),while the relative abundance of Desulfobacterota decreased in the low,medium and high-dose groups(P<0.05,P<0.01).The relative abundance of Chloroflexi decreased in the medium and high-dose groups(P<0.01).At the family level,the relative abundance of Muribaculaceae and Akkermansiaceae in the model group decreased(P<0.05,P<0.01);compared with the model group,the relative abundance of Muribaculaceae in Xiaoyao Pill combined with Guipi Decoction in high-dose group increased(P<0.05),and the relative abundance of Akkermansiaceae in low and medium-dose groups increased(P<0.05,P<0.01).In terms of genus level,the relative abundance of Akkermansia in the model group decreased(P<0.05),and the abundance of Ruminococcus decreased(P>0.05);compared with the model group,the relative abundance of Akkermansia in the Xiaoyao Pill combined with Guipi Decoction in the low and middle-dose groups increased(P<0.05,P<0.01),and the relative abundance of Rumenococcus in the low and high-dose groups increased(P<0.05);compared with fluoxetine group,the relative abundance of Rumenococcus in Xiaoyao Pill combined with Guipi Decoction medium-group was decreased(P<0.05).In terms of species level,the relative abundance of Akkermansia_muciniphila in the model group decreased(P<0.05);compared with the model group,the relative abundance of Akkermansia_muciniphila in the low and medium-dose groups of Xiaoyao Pill combined with Guipi Decoction increased(P<0.05,P<0.01).ConclusionXiaoyao Pill combined with Guipi Decoction can increase the weight of PPD model rats and improve depression-like behavior.The mechanism may be to increase the expression of5-HT,DA and BDNF in the hippocampus by regulating the structure and composition of the flora.
