Objective: By analyzing the healthy adult women,three groups of patients with breast fibroadenoma,breast cancer patients between intestinal flora differences in abundance and diversity,looking for the screening of the strong correlation between breast cancer and microbes,and explore the correlation between breast tumor and intestinal flora,for breast benign and malignant tumor provides some references for the condition assessment,treatment and follow-up.Materials and Mathods: Fresh morning stool samples from 36 healthy adult women,20 breast fibroadenoma patients,and 36 breast cancer patients were collected in the cache fluid,and stool samples from breast fibroadenoma patients and breast cancer patients were collected before surgery or primary adjuvant chemotherapy.Sequencing of 16 S r RNA V4 region was performed in 92 fecal samples.After processing the sequencing data,the diversity of OTU and α,the horizontal abundance of phyla and genus,and the metabolic pathways of intestinal flora were analyzed among the three groups.The abundance and diversity of intestinal flora among the three groups were analyzed and compared.The microflora data of the three groups in line with normal distribution were analyzed by univariate Anova test,and the non-normal distribution data were analyzed by Wilcoxon rank sum test,and the differences were statistically significant(P < 0.05).Results: A total of 8625625 high-quality sequences were obtained from 92 specimens in the three groups,and the average sequence length was about 303 bp.Under 97%similarity,the OTU were clustered into OTUS,and a total of 4352 OTUs were obtained.The number of OTUs shared among the three groups was 1342,and the sequencing coverage of all samples was 99.9%.Simpson index and overall α diversity were lower in breast cancer patients compared with healthy adult women and fibroadenoma patients(P<0.005).Similarly,the breast cancer group also showed a decrease in the number of sequenced fragments compared to the other two groups(P<0.01).PCA analysis showed that there were differences in the level of intestinal flora among the three groups.The abundance of Bacteroidetes in breast cancer patients was significantly lower than that in healthy women(P< 0.001)and fibroadenoma of breast(P<0.05).The abundance of Firmicutes in fibroadenoma patients was significantly higher than that in healthy adult women(P<0.01).The abundance of Firmicutes in breast cancer patients was higher than that in breast fibroadenoma patients,and the differences were statistically significant(P<0.05).Similar to the phylum level,PCA analysis showed differences in the composition of intestinal flora at the genus level among the three groups.At the genus level,the abundance of Ruminoccoccus increased gradually from the healthy adult female group to the fibroadenoma patient group and the breast cancer patient group.The abundance of Ruminoccoccus was highest in breast cancer patients and lowest in healthy adult women.Roseburia,Blaautia and Coprococcus also showed a similar trend.Prevotella and Bifidobacterium were statistically different only in the fibroadenoma group and the breast cancer group.The abundance of Prevotella and Bifidobacterium was lower in patients with breast cancer than in patients with breast fibroadenoma.In addition,KEGG function prediction analysis showed that there were significant differences in Biosynthesis and biodegradation of secondary metabolites between the breast cancer patients and the breast fibroadenoma patients.There was a significant difference in Ether lipid metabolism between breast cancer patients and healthy adult women.Similar to the comparative analysis of breast cancer patients and healthy adult women,there were significant differences in Ether lipid metabolism between breast fibroadenoma patients and healthy adult women.Conclusion: The diversity and species abundance of intestinal flora were different in breast cancer patients,breast fibroadenoma patients and healthy adult women,indicating that intestinal flora is related to the occurrence and development of benign and malignant breast tumors.This study provides a new idea for the clinical diagnosis and treatment of breast tumors.
