Purpose:To investigate the effect of triamcinolone acetonide(TA)on inhibiting scarring of drainage channel scarring after glaucoma filtration surgery in mice and its related mechanism.Methods:1.C57BL/6 mice were randomly divided into the normal control group,filtration surgery group and TA+ filtration surgery group.At different time points of modeling,the effects of TA on inflammation and collagen deposition in the bleb area of mice were studied by using hematoxylin-eosin(HE)staining and MASSON staining.At different time points of modeling,m RNA expressions of interleukin-6(IL-6),angiotensin II type 1a receptor(AT1a R),angiotensin II type 1b receptor(AT1b R),angiotensin II type 2 receptor(AT2R),matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9),connective tissue growth factor(CTGF)and transforming growth factor-β1(TGF-β1)in the tissue of filtration bleb area of mice in the normal control group,filtration surgery group and TA+filtration surgery group were detected by RT-PCR to study the anti-inflammatory,anti-collagen deposition and anti-fibrosis effects of TA in the bleb area after the operation.In order to study the anti-inflammatory and anti-fibrosis effects of TA in vivo,ELISA and Western blot were used to detect the expression of IL-6and CTGF proteins in the tissues of the filter bleb region of mice in the normal control group,filtration surgery group and TA+filtration surgery group.Results:1.In terms of the inflammatory response,compared with the normal control group,on the 1st,3rd and 7th day after surgery,the bulbar conjunctival area in the filter bleb area of mice in the filter surgery group was thickened,and a considerable number of inflammatory cells were infiltrated under the bulbar conjunctiva,while the infiltration of inflammatory cells under the bulbar conjunctiva in the TA+ filtration surgery group was less than that in the filtration surgery group.The expression of IL-6 m RNA and protein in the bleb area of mice peaked on the 1st postoperative day,and gradually decreased to the normal level on the 3rd,5th,7th and 14 th postoperative day.The expression of IL-6 m RNA and protein in the filtration region decreased in the TA+ filtration surgery group,and the difference was statistically significant.2.In terms of collagen deposition,compared with the normal control group,the collagen deposition area in the bleb region of mice in the filtration surgery group was significantly increased on the 28 th day after surgery,while the collagen deposition in the subconjunctival of the TA+filtration surgery group was less than that in the filtration surgery group.The m RNA expressions of AT1 a R,AT1 b R and AT2 R in the tissue of the filter bleb area of mice gradually increased on the 1st and 3rd day after surgery,and peaked between the 5th to 7th postoperative day,then gradually decreased to the normal level.The m RNA expression of AT1 a R and AT1 b R decreased in the TA+filtration surgery group,while the m RNA expression of AT2 R increased;on the 14 th day after surgery,the m RNA expressions of MMP-2 and MMP-9 in the bleb area of mice increased gradually and reached the peak on the 21 st to 28 th day.In the TA+filtration surgery group,the m RNA expression of MMP-2 and MMP-9 reduced and the change was statistically significant.3.In terms of fibrosis,the m RNA expressions of CTGF and TGF-β1 in the tissues of the filtered bleb region of mice increased gradually on the 14 th day after surgery,and peaked on the 21 st to 28 th day.The m RNA expression of CTGF and TGF-β1 decreased in the TA+ filtration surgery group.On the 14 th day after surgery,the expression of CTGF protein in the bleb region of mice increased progressively,peaking on the 21 st to 28 th day.The expression of CTGF protein decreased significantly in the TA+filtration surgery group.Conclusion:TA plays an anti-scar role in the bleb area of mice by decreasing the expression of the inflammatory factor IL-6 in the early stage and inhibiting collagen deposition and fibrosis in the middle and late stages. |
