Nonalcoholic fatty liver disease is a clinicopathological syndrome characterized by excessive deposition of fat in hepatocytes,excluding alcohol and other pathogenic factors that have definite damage to the liver.In recent years,the global incidence of NAFLD has remained high,which is mainly related to the unreasonable working,rest and eating habits of animals and humans.Taurine,a kind of sulfur-containing amino acid with simple structure,widely exists in human and animal bile.Taurine has a wide range of biological functions.Studies have shown that taurine can improve glucose and lipid metabolism in the liver of diabetic rats and reduce the triglyceride content of steatotic hepatocytes.However,its exact effect and mechanism on glucose and lipid metabolism of steatotic hepatocytes are not clear.In this experiment,the steatotic hepatocyte model was established by adding oleic acid(OA)and palmitic acid(PA)(OA: PA = 2:1)in vitro.At the same time,taurine intervention was used to detect the effect of taurine on glucose metabolism(glucose transport,glycolysis,glycogen synthesis,gluconeogenesis,glucose aerobic oxidation)and lipid metabolism(fatty acid metabolism,triglyceride metabolism,lipid droplet metabolism,cholesterol metabolism)steatotic hepatocytes.The purpose of this experiment is to clarify the regulatory effect of taurine on glucose and lipid metabolism of steatotic hepatocytes,which will provide a theoretical basis for the application of taurine in the prevention and treatment of NAFLD.The results showed that:1.Taurine intervention could significantly reduce the activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in steatotic hepatocytes,which indicates that taurine could improve the function of steatotic hepatocytes.2.Taurine intervention could significantly promote the content of intracellular glycogen and the m RNA expression of PFK,GYS2 in steatotic hepatocytes;it could significantly attenuate the glucose consumption,the content of lactic acid in culture medium and the m RNA expression of GLUT2,GK,GFAT2,MLXIPL,L-PK,GSK3β,CRTC2,PGC1α,FOXO1,PC,PEPCK,G6 Pase,DLAT,CS in steatotic hepatocytes.The results showed that taurine could improve the glucose metabolism of steatotic hepatocytes by inhibiting glucose transport,hexosamine pathway in glycolysis,glycogenolysis,gluconeogenesis and incomplete oxidative phosphorylation of glucose.3.Taurine intervention could significantly promote the content of intracellular HDL-C,the m RNA expression of PPARα,CPT1α,HADHB,ATGL,CGI58,MAGL,MTP,ABCG1,LDLR,CYP7A1 and the protein expression of PPARα,CPT1α,CYP7A1 in steatotic hepatocytes;it could significantly attenuate the content of intracellular TG,T-CHO,LDL-C,TBA,the m RNA expression of CD36,FATP5,L-FABP,SREBP1 c,PPARγ,LXRα,C/EBPα,ACC,FAS,SCD1,GPAM,DGAT2,PLIN2,PLIN3,SREBP2,HMGCR,ACAT2 and the protein expression of SREBP1 c,PPARγ,ACC,FAS,SREBP2,HMGCR,FXR.The results showed that taurine could improve the lipid metabolism of steatotic hepatocytes by inhibiting fatty acid uptake,de novo fatty acid synthesis,TG synthesis and lipid droplet differentiation,and promoting fatty acid oxidation,lipid droplet differentiation,apolipoprotein metabolism and cholesterol metabolism.In summary,taurine could improve the glucose and lipid metabolism of steatotic hepatocytes by inhibiting glucose transport,hexosamine pathway in glycolysis,glycogenolysis,gluconeogenesis,incomplete oxidative phosphorylation of glucose,lipid synthesis and promoting lipid decomposition.
