Objective: The whole body of deer is precious,but the utilization rate of deer bone,as a by-product of deer product processing,has been low.In order to solve the problem of the waste of deer bone resources,this study made the red deer bone into deer bone glue and deer bone degelatinatum,and conducted a quality analysis,and developed a series of products.Methods: 1.Ten batches of deer bone raw materials were collected to prepare deer bone colla and deer bone degelatinatum by high temperature cooking.Through single factor experiment and orthogonal experiment,the effects of the cooking temperature,cooking time and solid-liquid ratio on the yield rate of the deer bone colla and deer bone degelatinatum were investigated.The yield of the deer bone colla and deer bone degelatinatum was used to determine the best preparation process.2.The amino acid content was determined and the fingerprint was established by using the precolumn derivative method of phenyl isothiocyanate(PITC).The amount of calcium in deer bone degelatinatum was determined by atomic absorption spectrophotometry.3.(1)Through single factor experiment and orthogonal experiment,the effects of butter addition,peanut and marshmallow addition ratio and deer bone degelatinatum to milk powder addition ratio on the sensory quality of deer bone degelatinatum calcium supplement fudge were investigated.The best preparation process of deer bone degelatinatum calcium supplement fudge was finally determined by sensory evaluation.(2)Through single factor experiment and orthogonal experiments,the effects of the ratio of passion fruit raw juice and water,the ratio of proportion of white cool powder and fruit juice,the addition of xylitol and deer bone colla on the sensory quality of deer bone colla beautifying jelly were investigated.With the sensory evaluation,the best preparation process of deer bone colla beautifying jelly was finally determined.(3)The preparation process of deer bone degelatinatum calcium tablets was optimized by response surface method.The powder is directly compressed into tablets by using deer bone degelatinatum as raw material,microcrystalline cellulose,sodium carboxymethyl starch and magnesium stearate as auxiliary materials.Based on the single factor,the response surface method was used to optimize the fragility degree and disintegration time limit for determining the best preparation process.Results: 1.The optimal preparation process of deer bone colla and deer bone degelatinatum was determined as follows: temperature being 115℃,time being 16 h,and solid-liquid ratio being 1:6.2.The method of HPLC fingerprint and content determination of deer bone colla was established.This method has good accuracy,reproducibility and stability.The fingerprints of 10 batches of deer bone colla had good similarity to the control fingerprints,with a similarity of 0.990 to 1.000.In the deer bone colla control fingerprint,a total of 21 common peaks were identified and 16 chromatographic peaks were identified.Among them,the peak 1 is L-aspartate,peak 2 is L-glutamate,peak 3 is L-hydroxyproline,peak 4 is glycine,peak 5 is L-histidine,peak 7is L-threonine,peak 8 is L-alanine,peak 9 is L-proline,peak 13 is L-tyrosine,peak 14 is valine,peak 15 is L-methionine,peak 16 is L-isoleucine,peak 17 is leucine,peak 19 is L-phenylalanine,peak 20 is L-tryptophan,peak 21 is L-lysine.The main content determination components were L-hydroxyproline,glycine,L-alanine,and L-proline.The content determination method of deer bone colla is good and complies with the relevant regulations.The L-hydroxyproline content ranged from 7.91% to 9.71%,the glycine content ranged from 12.61% to 16.61%,the L-alanine content ranged from 4.70% to7.46%,and the L-proline content ranged from 8.94% to 12.46%.The calcium content determination method of deer bone degelatinatum is good and complies with relevant regulations.The calcium content of 10 batches of deer bone degelatinatum ranged from18.72% to 32.60%.3.(1)The best production process of deer bone degelatinatum calcium supplement fudge was as follows: the addition amount of butter being 7%,the addition ratio of peanut and marshmallow being 1:1,and the addition ratio of deer bone degelatinatum and milk powder being 1:10.The deer bone degelatinatum calcium supplement fudge tasted good,neither too soft nor too hard and moderately sweet,looked uniformly colorful,smelled nice,and never sticked to teeth.(2)The best preparation process of deer bone colla beautifying jelly was as follows: the addition ratio of raw passion fruit juice and water being 1:5,the addition ratio of white cool powder and fruit juice being 1:7,the addition amount of xylitol being 4%,and the addition amount of deer bone colla being 1%.The deer bone colla beautifying jelly prepared by this process had uniform color,tasted delicious,sweet and sour,cool and pleasant,and had the unique flavor of deer bone colla and passion fruit.(3)The best preparation process of deer bone degelatinatum calcium tablets was as follows: based on the addition of deer bone degelatinatum,the addition of microcrystalline cellulose being 45%,the addition of sodium carboxymethyl starch being 5%,and the addition of magnesium stearate being4.50%.The deer bone degelatinatum calcium tablets prepared by this process had a smooth surface and uniform color,and the friability and disintegration time were all in compliance with the regulations.Conclusion: In this study,through literature research and experimental results,the optimal preparation process of deer bone colla and deer bone degelatinatum was finally determined,which provided data reference for the product development of deer bone colla and deer bone degelatinatum.The amino acid content determination and fingerprint establishment by phenyl isothiocyanate(PITC)pre-column derivatization method,and the calcium content of deer bone degelatinatum by atomic absorption spectrophotometry,provide a theoretical basis for the quality evaluation of deer bone colla and deer bone degelatinatum.Through further product development of deer bone colla and deer bone degelatinatum,enrich the product types of deer bone products,laying a foundation for promoting the rational utilization and development of deer bone resources.
