| Research purposes:In this study,the aging rat models of D-galactose were established,the Ligustri Lucidi Fructus and Ecliptae Herba in Erzhi Pill were isolated and extracted,and the active parts were identified.The effects of the active parts of Erzhi Pill on the pathological state and relevant indexes in the tissue of model rats were observed,and various data were statistically analyzed to explore the anti-aging mechanism of the active parts of Erzhi Pill,so as to provide experimental basis to support for clinical anti-aging.Research methods:In this experiment,the Ligustri Lucidi Fructus and Ecliptae Herba in Erzhi Pill were separated and extracted by reflux method and macroporous resin adsorption method.The active parts of total flavonoids,total tri ter penes and total polysaccharides were extracted respectively,and the active parts were identified by spectrophotometry.Before the experiment,SD rats were randomly divided into 7 groups:blank group,model group,Erzhi Pill Decoction group,flavone group,triterpene group,polysaccharide group and active parts combination group.Eight weeks before the experiment,except the blank group,the other groups were injected intraperitoneally with 100mg/(kg·d)Dgalactose solution to establish the aging model of rats,and the blank group was given the same amount of normal saline.From the 9th week to the 14th week,the rats in the flavone group,triterpene group,polysaccharide group and active part combination group were intervened with the prepared corresponding experimental drugs.The decoction group was treated with Erzhi Pill Decoction with the same proportion and concentration,and the blank group and model group were perfused with normal saline.At the end of the 14th week,the rats were subjected to behavioral tests and recorded relevant data.After fasting and water deprivation,the rats were anesthetized with chloral hydrate the next day.The blood was taken quickly,the head was cut off,and the liver,brain and hippocampus tissues required for the experiment were extracted.The contents of SOD and MDA in rat liver,Mao and LPF in brain tissue and p 16 and p21 protein in hippocampus were measured by ELISA,and the expression of deletion mtDNA in hippocampus was detected by PCR.By detecting the above indicators,SPSS 25.0 statistical software is used to process and compare the data of each experimental group,to explore the anti-aging effects and mechanism of various active parts of Erzhi Pill on aging model rats and its correlation with oxygen free radical-mitochondrial pathway.Research results:①Erzhi Pill and active parts group can improve the general state and learning and memory ability of model rats.②Effects of SOD and MDA content in rats:the SOD content and MD A content in the model group decreased significantly,and the SOD content and MDA content in each administration group increased significantly,which was statistically significant.③Effects of Mao and LPF contents in rats:the MAO and LPF contents in the model group increased significantly,and the MAO and LPF contents in each administration group decreased significantly,which was statistically significant.④Effects of p16 and p21 protein content in rats:the p 16 and p21 protein content in the model group increased significantly,and the p16 and p21 protein content in each administration group decreased significantly,which was statistically significant.⑤Effects of mitochondrial DNA damage in rats:the deletion rate of mt DNA in the model group increased significantly,and the deletion rate of mt DNA in each administration group decreased significantly,which was statistically significant.Conclusion:1.Triterpenoids,flavonoids and polysaccharides were preliminarily isolated from the active parts of Erzhi Pill and identified.2.Through data analysis,it is concluded that the active parts of Erzhi Pill can improve the general state and learning and memory ability of aging model rats,regulate the contents of MDA and SOD in liver tissue,MAO and LPF in brain tissue,P16 and P21 protein contents in brain horse area and mtDNA deletion,indicating that the active part of Erzhi Pill has a certain anti-aging effect.3.Through data analysis and comprehensive evaluation,it is found that the active parts of Erzhi Pill has the same anti-aging effects as the decoction.The effects of combined administration group was better than that of decoction group in regulating monoamine oxidase,P16 protein and repairing mitochondrial DNA damage.4.The active parts of Erzhi Pill may play an anti-aging role by improving the activity of antioxidant enzymes,reducing the content of free radicals in the body,regulating the expression of aging protein and related gene pathways in hippocampus,improving brain neurotransmitter system and repairing the oxidative damage of mitochondrial DNA. |
