| Parkinson’s disease(PD)is the second most common neurodegenerative disease.α-Synuclein(α-Syn)is the main component of Lewy bodies and the pathogenic protein of PD which is the PD pathological marker.Therefore,the PD is also referred to as α-Synuclein disease.The traditional PD models(neurotoxin-induced or gene mutation)have the disadvantages including rapid disease progression,severe damage,and difficulty in reproducing Lewy pathology.Compared with rodents,non-human primates brain structure,physiological and biochemical mechanisms,behavioral performance and genetic background are more similar to humans,which can better simulate the occurrence and development of human diseases.Therefore,using α-Syn induces the PD model of the cynomolgus monkey that is closer to PD pathogenesis,which will provide reference for the research of pathogenic mechanism and treatment of PD.Therefore,the purpose of this study: 1)Collect α-Syn induces the PD model of monkey data including non-motor symptoms and motor symptoms by behavior testing,and assess the similarity of symptoms of PD model and PD patients.2)Through imaging detection,use non-invasive DAT-SPECT to detect the dopaminergic neurons of PD model,and obtain imaging data of different time periods,which assessing the injury process of dopaminergic neurons in the PD model of monkey.3)Pathology analysis of PD monkey model substantia nigra to verify the success of the model construction.Our results found that α-Syn monkey model appears similar PD patients with fine motor dysfunction,autonomous activity disorders and tremors,through fine motor function test,PD symptom score,autonomous activity distance detection,bromocriptine treatment experiment and aponemorphine induced rotation experiment in behavioral testing.Sugar water preference experiment and sleep disorder experiments have found that non-motor symptoms of α-Syn monkey model appears similar to PD patients including depression.In imaging detection,DAT-SPECT detection shows that the α-Syn monkey model extends to exhibit a dopaminergic neuronal impairment of similar PD patients.This study evaluated the similarity of α-Syn monkey model and PD patients through behavioral and imaging,and we found dopamine neurons of substantia nigra in PD monkey model show the aggregation of phosphate acidization α-Syn and the reduction of the number of dopamine neurons by tissue pathological analysis.Therefore,the establishment of α-Syn-induced PD model of cynomolgus monkey has great similarities in behavioral and imaging,which is of great significance for studying the PD pathogenic process and pathogenesis. |
