Liver injury is a fatal disease that affects the quality of human life,and its pathogenesis is very complex.The study of its pathogenesis and the mechanism of drug action is helpful for doctors to improve treatment plan.In addition,it can enhance the understanding of the selected small molecules of traditonal Chinese medicine and realize the extension of the small molecules to treat other diseases affected by the related signaling pathways in humansThe purpose of this study was to construct different liver injury mouse models under various conditions,and then verify the pharmacodynamic effects of various Chinese traditional medicine active ingredients on the liver injury mouse models constructed,and verify the mechanism of action of effective Chinese traditional medicine ingredients on liver injury based on this exploration experiment.The hepatic injury model of mice was induced by carbon tetrachloride,Concanavalin A(Con A),methionine choline deficiency and dietary methionine choline deficiency.After the liver injury model was successfully established by blood biochemistry,enzyme-linked immunosorbent assay,pathological section and other physiological indicators and pathological change confirmation,the efficacy test was carried out on the established model animals.Use of active ingredients including celastrol and ginsenosides Rh1,ginsenosides Rh1,glycyrrhizin the active components of traditional Chinese medicine treatment of building mice eventually again through the blood biochemistry and the measurement of enzyme-linked immune adsorbent,pathological,and flow cytometry method confirmed that the traditional Chinese medicine composition for the improvement of liver injury and explore the mechanism of action.Through animal experiments and found the above using the four Chinese native medicine ingredient of celastrol on the use of Con A induced liver injury in mice model has relatively significant improvements,and other traditional Chinese medicine composition can be respectively for one of blood biochemical and pathological indicators have a certain effect,but the overall improve the effect of a complete cure liver damage does not function.In vitro experiments,it was found that celastrol up-regulated the number of myeloid immune cells differentiating into Ly6 low subtypes in the liver.Combined with the results of in vivo and in vitro experiments found in celastrol of Con A mechanism may be because the main function of the liver damage induced by Con A induced liver injury,cause immune cells in liver,prompting form autoimmune liver damage,the liver damage in the process of intrahepatic macrophage apoptosis after there will be other immune cells in the blood and bone marrow to the liver,And injection of celastrol of the experimental flow cytometry results showed that the ly6 Clow subtypes of immune cells from an increase in the number of element that celastrol may be able to promote to the liver of myeloid immune cells to differentiate into ly6 Clow subtype,but studies have found that the subtypes of immune cells have certain improve fiber degradation and liver damage repair function,Therefore,it is speculated that celastrol mainly acts on the signal pathways of related immune cell differentiation and can achieve the intervention effect on liver injury by promoting the differentiation of immune cells into ly6 Clow subtypes.
