Objective:To investigate the relationship between BRCA1/2 and E-cad expression and clinicopathological features in mammary invasive carcinoma tissues.Methods:1.The clinical and pathological data of 431 patients with breast invasive carcinoma diagnosed in Shanxi Cancer Hospital from January 2019 to December2019 were retrospectively analyzed,and the E-cad protein and NGS method were used to detect BRCA1/2 gene by immunohistochemical method.The clinicopathological features in mammary invasive carcinoma tissues and their correlations were analyzed by X^2 test and Spearman.2.GEPIA analyzes the expression and correlation of BRCA1 and BRCA2 genes in breast invasive carcinoma tissues in the TCGA database.3.UALCAN analyzes the expression and clinical characteristics of BRCA1 and BRCA2 in breast invasive cancer tissues in the TCGA database.Results:1.The expression of E-cad protein was inversely correlated with the expression of PR(r=-0.113,P=0.019<0.05);the expression of E-cad protein was positively correlated with PTNM staging(r=0.010,P=0.043<0.05);the expression of BRCA1/2gene was positively correlated with subarm lymph node metastasis(r=0.105,P=0.029<0.05);the expression of BRCA1/2 gene was negatively correlated with the expression of Ki-67(r=-0.143,P=0.003<0.05);BRCA1/2 mutation rate was inversely correlated with E-cad expression(r=-0.098,P=0.041<0.05).2.The relative expression of m RNA of BRCA1 and BRCA2 increased significantly in mammary invasive carcinoma tissues,and the difference was statistically significant;the expression levels of BRCA1 and BRCA2 were significantly positively correlated.3.BRCA1 m RNA expression in breast invasive carcinoma tissues was significantly higher than that in normal tissues(P<0.001);BRCA1 m RNA expression in male patients was higher than in female patients(all P<0.05);BRCA1m RNA expression in N3 primary tumors was lower than other stages(all P<0.05),and N2 was lower than that in N1 primary tumors(P<0.05);however,there were no significant differences in N0 and N1,N0 and N2,N0 and N3,N1 and N2 BRCA1 m RNA expressions(P>0.05).There were no significant differences in BRCA1 m RNA expression in primary tumors of patients with different ages,molecular typings,and different menopausal states(P>0.05).BRCA2 m RNA expression in breast invasive carcinoma tissues was significantly higher than that in normal tissues(P<0.001);the expression of BRCA2 m RNA in N3 primary tumors was lower than that of other stages(all P<0.05),and the expression of BRCA2 m RNA in N0 and N1 was lower than that of N2 primary tumors(P<0.05);however,there were no significant differences in BRCA2 m RNA expression between N0 and N1,N0 and N2,and N1 and N2(P>0.05);BRCA2m RNA expression in TNBC patients was higher than that in Luminal patients(P<0.05).There were no significant differences in BRCA2 m RNA expression in primary tumors of patients of different ages,sexes and menopausal states(P>0.05).Conclusions:1.BRCA1/2 and E-cad expression in mammary invasive carcinoma tissues were related to PR,presence or absence of axillary lymph node metastasis,PTNM staging,and Ki-67 expression;and BRCA1/2 was negatively correlated with E-cad.2.The relative expression of m RNA of BRCA1 and BRCA2 increased significantly in breast invasive carcinoma tissues,and the expression levels of BRCA1 and BRCA2 were significantly positively correlated.3.BRCA1 m RNA expression in mammary invasive carcinoma tissues is significantly higher than that of normal tissues;BRCA1 m RNA expression correlated with patient sex and axillary lymph node metastases,and was not statistically significant with age,molecular typing,and menopausal status.BRCA2 m RNA expression in breast invasive carcinoma tissues was significantly higher than in normal tissues;BRCA2 m RNA expression was associated with axillary lymph node metastasis and molecular typing,and was not statistically significant with age,sex,and menopausal status.In summary,BRCA1 is positively correlated with BRCA2;BRCA1/2 is negatively correlated with E-cad in mammary invasive carcinoma,both of which are associated with clinical pathological features.It can provide reference or verification for the study of relevant mechanisms,and provide direction for the discovery of new biomarkers and targeted therapies.
