Objective:To develop the preparation technology of total flavonoids liposomes from hawthorn leaves in order to improve the bioavailability,on this basis,conduct a preliminary study on the formulation of traditional Chinese diet dietary preparations with total flavonoids from hawthorn leaves as the main components.Methods:1、Evaluating the content and antioxidant activityThe total flavonoids from hawthorn leaves were extracted and purified using ethanol and D101 macroporous resin;the flavonoid contents was measured by chromogenic;we then performed DPPH,ABTS and hydroxyl free radical assays to monitor the antioxidant potential of total flavonoids.2、Optimizing of preparation process and quality evaluation of hawthorn leaves total flavonoid liposomes,and developing a dietary therapy product with hawthorn leaves total flavonoid liposome as main componentThe total flavonoids of hawthorn leaves extracted by liposome technology and film dispersion-ultrasonic method were used as raw materials,and the encapsulation degree is used as an indicator.Planning and optimization of the preparation process.The optimal preparation method was used to prepare the liposomes,and the physicochemical properties of the liposomes were evaluated by appearance,particle size,zeta potential and transmission electron microscope(TEM)images.On this basis,by referring to relevant literature and invention patents,we explored the dietary therapy preparations of traditional Chinese medicine from three aspects:composition,formula and preparation process of formula.Results:1、The content of total flavonoids in hawthorn leaves was(77.55±2.33)mg/g.The half maximal inhibitory concentration(IC50)values of total flavonoids from hawthorn leaves on DPPH and ABTS free radical scavenging were(91.63±1.75)and(64.34±4.53)μg/m L,respectively;compared with ascorbic acid,the relative antioxidant activity(RAA)was 40.71%and 76.64%;meanwhile,hydroxyl free radical clearance rate was low,all three were lower than ascorbic acid under the same mass concentration.2、The optimal preparation process of liposomes was as follows:lecithin to cholesterol mass ratio 5:1,lecithin concentration 30 mg/ml,lecithin and hawthorn leaves total flavonoids mass 10:1,and the addition amount of Tween 80 accounts for 10%of the mass of lecithin.The liposomes prepared under this condition were light yellow,and the encapsulation efficiency,particle size and Zeta potential were(73.25±2.13)%,(132.2±2.85)nm,(-9.29±0.34)m V,respectively.The storage stability results showed that the stability was better at 4℃,which could efficiently utilize hawthorn leaves total flavonoids.Conclusion:Hawthorn leaves collected in Jinnan area of Shanxi have a high abundance of total flavonoids and high antioxidant activity;total flavonoid liposomes from hawthorn leaves prepared by the thin-film hydration method,is stable and feasible,which can improve the bioavailability of total flavonoids.In the later stage,the research group will use the total flavonoids of hawthorn leaves,poria,pueraria,curcumin and steviol glycosides as compatibility to study the preparation process of traditional Chinese diet dietary products.
