Objective: Bioinformatics analysis was carried out through GEO and Oncomine databases.Combined with the verification of experimental methods,the differential expression of GATA3,BCL11 B and CDKN2 A in peripheral T-cell lymphoma was explored,and the differences in the expression of GATA3,BCL11 B and CDKN2 A in PTCL and reactive proliferative lymph nodes,and the significance of clinical pathological characteristics and prognosis were analyzed to guide the accurate classification,early diagnosis and individualized treatment of PTCL.Methods: 1.The expression of GATA3,BCL11 B and CDKN2 A m RNA in PTCL tissues and their correlation were analyzed by GEO and Oncomine databases.2.The clinical data and paraffin-embedded specimens of 123 patients with PTCL diagnosed in Shanxi Cancer Hospital from January 2010 to May 2020(with complete clinical course data and follow-up records)were collected as the experimental group,and 40 cases of reactive hyperplastic lymph nodes were randomly selected as the control group.Tissue microarray was made.The expression levels of GATA3,bcl11 b and CDKN2 A proteins in the experimental group and the control group were detected by immunohistochemical staining.3.The expression of GATA3,BCL11 B and CDKN2 A m RNA in 80 cases of PTCL and 40 cases of LH were detected by reverse transcriptase polymerase chain reaction.4.Statistical software SPSS25.0 was used to analyze the differential expression of GATA3,BCL11 B and CDKN2 A protein and m RNA in the experimental group and the control group,and to analyze the correlation between the three,and the relationship between their expression and the pathological factors of PTCL disease.Drawing using Graph Pad Prism 8.0.Results: 1.Results of database analysis : According to the data set GSE132550 in GEO database,the transcription levels of GATA3 and BCL11 B in PTCL cases were lower than those in normal T cells,while the transcription level of CDKN2 A in PTCL cases was higher than that in normal T cells(P < 0.05).Oncomine database showed that the expression of GATA3 and BCL11 B was correlated.2.IHC result:the positive rates of GATA3 and BCL11 B were both located in the nucleus,and the positive expression rates in PTCL tissues were 60.98%(75 / 123)and 56.10 %(69 / 123),respectively,while the positive rates in LH tissues were 85.00 %(34 / 40)and 75.00%(30 / 40),respectively.The differences in the expression of GATA3 and BCL11 B proteins between the two tissues were statistically significant(P < 0.05).CDKN2 A protein was accumulated in cytoplasm / nucleus,and the positive rate in PTCL tissue was 79.67 %(98 / 123),while that in LH tissue of the control group was 70.00 %(28 / 40),with no significant difference(P > 0.05).3.RT-PCR result:The relative expression levels of GATA3 and BCL11 B m RNA in 80 PTCL patients were lower than those in the control group(0.678 vs 1.855,0.702 vs 1.832,respectively),and the difference was statistically significant(P < 0.05).CDKN2 A m RNA relative expression was slightly higher than the control group(1.113 vs 1.081),the difference was not statistically significant.4.Relationship between GATA3,BCL11 B and CDKN2 A expression and clinicopathological factors : GATA3 expression was correlated with Ann Arbor stage and pathological type,and CDKN2 A expression was correlated with pathological type(all P < 0.05).5.The correlation between the relative expression levels of GATA3,BCL11 B and CDKN2 A m RNA and protein expression levels in PTCL tissues : The results of correlation analysis showed that there was no significant correlation between the relative expression levels of GATA3,BCL11 B and CDKN2 A m RNA and protein expression levels in the experimental group(P > 0.05).6.Correlation of GATA3,BCL11 B and CDKN2 A expression in PTCL : Spearman correlation analysis showed that the expression of BCL11 B protein in PTCL tissue was significantly positively correlated with that of GATA3 protein(r = 0.357,P < 0.01).7.Prognostic significance of GATA3,BCL11 B and CDKN2 A expression in PTCL : in the low expression group of GATA3 m RNA,the overall survival time was slightly shorter in the high expression group;in the low expression group of BCL11 B gene,the overall survival time was shorter than that in the high expression group(all P < 0.05).The total survival time of BCL111b-/ GATA3-group was shorter than that of BCL111 b + / GATA3 + group(P < 0.05).BCL11 B protein is an independent risk prognostic factor for PTCL.Conclusion: Through database analysis and experimental verification,it is known that BCL11 B and GATA3 are differentially expressed in PTCL and LH,and affect the prognosis of the disease.There was no difference in CDKN2 A expression between PTCL and LH.BCL11 B and GATA3 may be involved in the occurrence and progression of diseases,and are expected to become new biomarkers for predicting prognosis and treating diseases.
