Objective:To investigate the expression of SRGN in cervical and normal cervical tissues and its correlation with epithelial interstitial transformation.Methods:The differential expression of SRGN in different tumors and its survival and prognosis in cervical cancer were analyzed by GEPIA2 database mining.Secondly,the differential expression of SRGN in cervical cancer tissues and normal cervical tissues was analyzed by GEO database mining data set GSE9750.TIMER2 database analyzes the correlation between SRGN and CAFs.Paraffin sections of 70 patients with cervical cancer and 30 patients with hysterectomy without cancer were collected,as well as clinicopathological datas.Hematoxylin-eosin staining(HE)was conducted on cervical cancer tissue and normal cervical tissue,and different pathological conditions were analyzed.The expression of SRGN,E-cadherin and Vimentin in cervical and normal cervical tissues and their relationship with clinicopathological parameters were analyzed by immunohistochemical staining in paraffin sections of 100 patients.Immunohistofluorescence assay was used to detect the expression of the three proteins in cervical cancer tissues with lymph node metastasis and cervical cancer tissues without lymph node metastasis.Western-blot(Wb)was used to detect the expression of SRGN,E-cadherin and Vimentin.Results:1.Bioinformatics analysis showed that SRGN had significant effects on the growth of Esophageal cancer(ESCA),GBM,Acute Myeloid Leukemia(LAML),Brain Lower Grade Glioma(LGG),Ovarian serous cystadenocarcinoma(OV),Pancreatic adenocarcinoma PAAD),Testicular Germ Cell Tumors(TGCT)and Skin Melanoma(SKCM)tumor tissues showed significantly higher gene expression levels than the corresponding control group(P < 0.05).The gene expression level of SRGN in cervical cancer tissue was higher than that in normal cervical tissue(P < 0.05).The expression level of SRGN in cervical cancer was positively correlated with the expression level of CAFs invasion(P < 0.05).2.The gene expression level of SRGN in cervical cancer tissues was higher than that in normal cervical tissues(P < 0.05).Immunohistochemical test results showed that SRGN and Vimentin expression levels in cervical cancer tissues were higher than that in normal cervical tissues(P < 0.001),while E-cadherin expression levels were lower than that in normal cervical tissues(P < 0.01).Immunofluorescence experiment showed that the expression levels of SRGN and Vimentin in cervical cancer tissues with lymph node metastasis were higher than those in cervical tissues without lymph node metastasis,while the expression levels of E-cadherin were opposite(P < 0.05).SRGN was negatively correlated with e-cadherin(r=-0.44,P < 0.001)and positively correlated with Vimentin(r=0.40,P < 0.001).The relationship between SRGN and clinicopathological parameters was analyzed.It was found that SRGN had no correlation with age,differentiation degree and vascular tumor thrombin of cervical cancer patients,but was closely related with lymph node metastasis and TNM stage.WB results were consistent with immunohistochemical results.Conclusion:SRGN is highly expressed in cervical cancer tissues,and may regulate and participate in the development of EMT and promote the invasion and metastasis of cervical cancer.At the same time,SRGN is closely related to TNM stage and lymphnode metastasis in patients with cervical cancer,and can be used as a clinical prognosis assessment of cervica... |
