Objective:To study the effect of Modified-buwang-powder on the cognitive function of vascular dementia rats and explore the effect of Modified-buwang-powder on cholinergic system.Methods:60 healthy male SD rats(200g±20g)were randomly divided into 6 groups,respectively,were marked as control group,the Va D model group(model group),donepezil group,lowdose Modified-buwang-powder group(hereinafter referred to as the lower dose group),and medium-does Modified-buwang-powder group(hereinafter referred to as the medium dose group),high-dose Modified-buwang-powder group(hereinafter referred to as high dose group).Bilateral carotid artery ligation was used to prepare the Va D model.In the sham operation group,only the carotid artery was separated without ligation.Ten days after surgery,the treatment groups were given 0.05g/m L,0.15 g/m L and 0.45 g/m L of Modifiedbuwang-powder solution by gavage,respectively.Donepezil group was given donepezil hydrochloride solution at 0.5mg/kg.Model group and sham operation group were given the same dose of normal saline intragastric.Morris water maze and open field test were used to evaluate the learning and memory ability of rats after 30 days of intragastric administration.Acetylcholine Acetyltransferase(Ch AT)and Acetylcholinesterase(ACh E)were detected by Western blot.Results:1.Behavioral experiment results:(1)Morris Water Maze: In the positioning navigation experiment,the escape latency of rats in each group decreased with the extension of training days.In the space exploration experiment,compared with the model group,there was no statistical difference in the lowdose group(27.2±2.71 VS 23.78±3.14,P > 0.05),while the percentage of dwell time in the original platform quadrant of the medium-dose and high-dose groups was significantly prolonged(28.58±2.57 VS 23.78±3.14,P > 0.05).P< 0.05;32.69±2.72 VS 23.78±3.14,P<0.01).In the visual platform experiment,there were no significant differences in the time required for rats to climb up the platform and swimming speed among all groups(P > 0.05).(2)Open field experiment: compared with the model group,the penetration times of low-dose,medium-dose and high-dose groups increased,and the difference was statistically significant(80±10.28 VS 61.125±16.5,P< 0.01;92.75±4.95 VS 61.125±16.5,P< 0.01;111.75 ± 7.8 VS 61.125 ±16.5,P<0.01),and with the increase of the concentration of Modified-buwang-powder,the number of penetration also increased.There was no significant difference in the number of perforations between the high-dose group and the Donepezil group(111.75±7.8 VS 124.375±6.32,P > 0.05).2.Western Blot results:(1)Compared with model group,Ch AT expression level in hippocampus was increased in low,medium and high dose groups with statistical significance(0.622±0.029 VS0.442±0.058,P< 0.01;0.715±0.035 VS 0.442±0.058,P<0.01;0.809±0.028 VS 0.442±0.058,P<0.01).There was no significant difference in Ch AT content between the high-dose group and the sham group(0.809±0.028 VS 0.84±0.033,P>0.05).(2)Compared with model group,the content of ACh E in hippocampus of rats in medium-dose and high-dose groups was decreased(0.76±0.102 VS 0.888±0.063,P<0.05;0.648±0.104 VS 0.888±0.063,P<0.01);Compared with donepezil group,there was no difference in the content of ACh E in hippocampus of rats in high-dose group(0.648±0.104 VS 0.725±0.053,P>0.05);There was no significant difference between the high-dose group and the sham group(0.648±0.104 VS 0.598±0.033,P>0.05).Conclusion:1.The addition of Modified-buwang-powder may improve the learning and memory ability of Va D model rats in a dose-dependent manner.2.The expression level of Ch AT and ACh E in the hippocampus of Va D model rats could be dose-dependently increased by Modified-buwang-powder. |
