Objective:To investigate effects of the fibrin-derived peptide Bβ15-42 on cell apoptosis after renal ischemia reperfusion injury in rats.Methods:The SD rats were assigned into the sham operation group(Sham group),renal ischemia reperfusion group(I/R group)and FgBβ15-42 treatment group(Fg+I/R group)by the random number table method.Renal I/R model was established by clipping bilateral renal arteries for 60 minutes and then restoring blood of bilateral renal arteries.Blood and bilateral kidney samples were collected after reperfusion for 24 hours.Scr and BUN were tested to valuate renal function;renal histopathological injury was observed by HE staining and the degree of injury was valuated by semi-quantitative analysis;cell apoptosis was observed by TUNEL staining;the protein expressions of Caspase-3,Bax and Bcl-2 were determined by immunohistochemistry and Western blot.Results:(1)Compared with Sham group,Scr and BUN were obviously increased in I/R group(P<0.05);some tubular epithelial cells swelled,brush border lost,some epithelial cells became necrotic and shed,tubular lumen got enlarged,inflammatory cell infiltrated in renal interstitium and the score of renal injury was increased in I/R group(P<0.05);cell apoptosis rate was increased in I/R group(P<0.05);the expression of Caspase-3 and Bax protein were increased,the expression of Bcl-2 protein and the ratio of Bcl-2/Bax were decreased in I/R group(P<0.05).(2)Compared with I/R group,Scr and BUN were obviously decreased in Fg+I/R group(P<0.05);renal histopathological injury got lighter and the score of injury was decreased in Fg+I/R group(P<0.05);cell apoptosis rate was decreased in Fg+I/R group(P<0.05);the expression of Caspase-3 and Bax protein were decreased,the expression of Bcl-2 protein and the ratio of Bcl-2/Bax were increased in Fg+I/R group(P<0.05).Conclusion:(1)The FgBβ15-42 peptide protected renal ischemia reperfusion injury;(2)Renal protection of FgBβ15-42 peptide was associated with decreasing Bcl-2/Bax-mediated mitochondrial pathway cell apoptosis.
