Objective: Lung cancer is currently the leading cause of death from malignancy worldwide,and lung adenocarcinoma(LUAD)is the most common non-small cell lung cancer.Immune related genes(IRGs)play an important role in the development and progression of lung cancer and are associated with prognosis.In this study,a prognostic model of immune related genes in lung adenocarcinoma was constructed based on bioinformatics methods to evaluate the prognosis of lung adenocarcinoma patients and the benefit of immunotherapy.Methods: Differentially expressed genes between 535 LUAD tissues and 59 adjacent lung samples were obtained from TCGA database,and immune genes were obtained from Imm Port and Innatedb database.WGCNA co-expression network construction,univariate COX survival analysis,and multivariate COX analysis were performed on immune related differential genes to obtain immune related prognostic model genes and their risk coefficients.They were divided into high and low risk groups according to the median value of risk coefficients,and the chip GSE72094 gene set was selected from the geo database to validate the model reliability by using time-dependent ROC curve and univariate and multivariate independent prognostic analysis.Finally,we analyzed the benefit of immunotherapy in two groups by TIDE and proved the superiority of the model compared with TIDE or TIS models.Results: An immune-related prognostic gene model consisting of nine immune genes(PTGDS、S100A16、ANGPTL4、LIFR、SHC3、TRIM6、PLK1、C7、C6)was constructed,which could stratify patients into high-risk group and low-risk group.Compared with the low-risk group,the high-risk group showed significantly worse survival in the experimental and test groups(P < 0.001).The model was proved to be reliable by time-dependent ROC curve,univariate and multivariate COX regression analysis.Finally,the TIDE score showed that the high-risk group was sensitive to immunotherapy,and the model was superior compared with the TIDE and TIS models.Conclusion: The lung adenocarcinoma immune-related gene prognosis model constructed in this study can effectively predict the prognosis of patients,and is related to the evaluation of the benefits of immunotherapy,providing a new idea for the diagnosis and treatment of lung adenocarcinoma.
