| Objective:To explore the prognostic value of ferroptosis-related lncRNA in hepatocellular carcinoma patients and their potential value in tumor microenvironment.LncRNA risk scoring system and nomogram were constructed to predict patient survival.Methods:Download hepatocellular carcinoma patient data from TCGA database.Download the iron death related cause list from the Ferr Db website database.ferroptosis-related lncRNA were screened by constructing iron death-related m RNA-lncRNA co-expression network.The prognostic model and risk scoring system of ferroptosis-related lncRNA were established by univariate COX regression,LASOO regression and multivariate COX regression.K-m survival curve and ROC curve were drawn to evaluate its predictive ability,to explore the sensitivity and specificity of risk score and different clinical features for predicting survival and prognosis of HCC patients,and to compare the predictive ability of different clinicopathological features and risk score.The factors affecting the survival and prognosis of HCC patients were used to construct the nomogram,and the accuracy of the prognosis of HCC patients was evaluated by correction curve,C index and ROC curve.Results:Five prognostic models of ferroptosis-related lncRNA were obtained,including AL049840.4,ZFPM2-AS1,AC074117.1,AC099850.3 and AL031985.3.The results of risk score and TNM stage showed that the model had good predictive ability.KEGG pathway results showed that the high-risk group was enriched in classic tumor-related pathways.The prognostic model is related to tumor immune cell infiltration,immune function and expression of immune checkpoint genes.Conclusion:The prognostic model of ferroptosis-related lncRNA constructed in this study has certain accuracy and reliability for predicting the survival prognosis of hepatocyte patients,providing clues for the interaction between ferroptosis and HCC immunity.Targeting iron death combined with immune checkpoint inhibitors is expected to provide a new perspective for HCC treatment. |
