Objective:Bioinformatics methods were used to analyze ferroptosis-related subtypes in patients with clear cell renal cell carcinoma(ccRCC),choose ferroptosis-related genes significantly related to prognosis,and establishment of a prognostic nomogram.Methods:The transcriptome data and clinical data of 72 paracancer normal tissues and 526 ccRCC patients with clinical data were downloaded from The Cancer Genome Atlas database.Partitioning Around Medoids algorithm was used to determine the number of subtypes.Gene Ontology functional enrichment analysis and Kyoto encyclopedia of genes and genomes functional enrichment analysis were used to explore potential functions and pathways.Univariate Cox analysis,Kaplan-Meier survival curve analysis,least absolute shrinkage and selection operator regression analysis and multivariate Cox analysis were used to choose the ferroptosis-related genes of ccRCC,and the nomogram was establishment.Results:Four ferroptosis-related subtypes of ccRCC were identified and their clinical features and survival characteristics were analyzed.Enrichment analysis showed that subtype signature genes were closely related to cellular immunity and tumor microenvironment.Ten ccRCC ferroptosis-related genes(HMOX1,MT1 G,CHAC1,LURAP1 L,CXCL2,RRM2,TRIB3,DUSP1,GABARAPL1,ALOX12B)were choosed.A nomogram containing age,M staging,pathological staging,tissue grading and risk score was established,and the areas under the ROC curve of the nomogram in 1,3 and 5 years were 0.858,0.809 and 0.774,respectively.Conclusion:Four ferroptosis-related subtypes of ccRCC were identified,and the subtypes were related with clinical prognosis.The nomogram has good predictive ability for the prognosis of patients with ccRCC,and the ferroptosis-related genes of ccRCC in the model can be used as targets for studying the pathogenesis and targeted therapy of ccRCC.
