Objective: To observe the neuroprotective effects of remazolam(RZ)on subarachnoid hemorrhage(SAH)rats.The neurological dysfunction score,brain histopathology,brain water content,interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α),S100 β in SAH rats were measured.To study the neuroprotective effect of RZ on SAH rat.Methods: Forty five male SD rats weighing 280-350 g were randomly divided into three groups.The SAH model was established by the modified occipital cistern secondary blood injection method.The Sham operation group(sham group)were injected with 0.3ml normal saline into the occipital cistern,while the subarachnoid hemorrhage group(SAH group)was injected with 0.3ml non anticoagulant autologous arterial blood into the occipital cistern,then he subarachnoid hemorrhage pump group(SAH+RZ group)was injected with 0.3ml non anticoagulant autologous arterial blood into the occipital cistern.After successful modeling,SAH+RZ group was given RZ 2mg / h and pumped for 2h,while SAH group was given normal saline 1.5ml/h and pumped for 2h.24 h after operation,the rats were scored with the improved Garcia score,the pathomorphology was observed by HE staining,the water content of brain tissue was measured by dry wet weight method,and the IL-6 and TNF-α in brain tissue samples were detected by Western blot(WB)and S100 β Relative Expression.Results: Compared with Sham group,the score of neurological dysfunction in SAH group decreased(P< 0.01),and that in SAH+RZ group increased(P< 0.01);HE staining showed that the number of neurons in SAH group decreased compared with sham group,and the number of neurons in SAH + RZ group increased compared with SAH group;The results of brain water content showed that compared with sham group,the brain water content of SAH group increased(P< 0.01),and that of SAH + RZ group decreased(P< 0.01);WB method was used to detect the expression of related inflammatory factor protein.It was found that compared with sham group,SAH group had higher TNF-α,IL-6 and S100 β Relative Expression.The expression of TNF-α,IL-6 and S100β in SAH + RZ group were significantly higher than those in SAH group(P<0.01),as the expression was significantly decreased(P< 0.01).Conclusion: Remazolam can play a neuroprotective role by improving the neurological dysfunction of rats with subarachnoid hemorrhage and reducing the pathological changes,brain edema and inflammatory reaction of brain tissue in rats with subarachnoid hemorrhage.
