Objective:In this study,we explore the modulation effect of postbiotics on the oral microbiota by randomised controlled trials on the population of postbiotics adjuvant therapy of caries filling for 14 days.Methods:Ninety volunteers were recruited and divided into group A(postbiotics adjuvant therapy of caries filling group,n=41),group B(caries filling therapy group,n=40),and group C(oral health group,n=9).All volunteers collected oral saliva samples at 0 and14 days after therapy,and the defined groups were A_0,A_14,B_0,B_14,C_0,C_14.The collected oral saliva samples were sequenced by 16S rRNA,and the sequencing results were analyzed by bioinformatics.Results:1.The oral microbiota diversity analysis of postbiotics adjuvant therapy of caries filling,caries filling therapy and oral health population showed that,unlike group C,the microbiota alpha diversity of group A and B samples on day 14 was significantly lower than that on day 0,with group A showing a more significant decrease in microbiota alpha diversity than group B(P<0.001).Meanwhile,after 14 days of therapy,the microbiota richness of group A_14 was significantly higher than that of group B_14,and the microbiota was more stable(P<0.01).Beta diversity analysis showed that the decrease in the diversity of the microbiota in groups A and B clearly changed the structure of the microbiota(A0 vs A14,P=0.004,R~2=0.027;B0 vs B14,P=0.005,R~2=0.025).Although there was no significant difference in the colony structure of the A_14,B_14,and C_14 groups,the microbiota structure of the A_14group was closer to that of the C_14 group than that of the B_14 group(A_14 vs C_14,P=0.602,R~2=0.019;B_14 vs C_14,P=0.299,R~2=0.024).2.The analysis of oral microbiota species differences between postbiotics adjuvant therapy of caries filling,caries filling therapy and oral health population showed that after 14 days of therapy,groups A and B changed from the microbiota dominated by caries-associated Firmicutes and Streptococcus to the microbiota dominated by Proteobacteria and Pseudomonas(LDA>4,P<0.001).Compared with the C_14 group,the A_14 group was enriched with health-related Proteobacteria and Neisseria(LDA>4,P<0.001),while the B_14 group was significantly enriched with dysbiosis-related Cyanobacteria(LDA>4,P<0.001).Meanwhile,the A_14 group was enriched with health-related Neisseria.compared with the B_14 group(LDA>4,P<0.001),while the B_14 group was enriched with caries-associated Firmicutes and Streptococcus(LDA>4,P<0.001).Conclusion:Caries filling therapy could partially restore the imbalanced oral microbiota.Adjunctive postbiotic tablets regulate the oral microbiota after caries filling treatment by increasing the stability of microbiota and shifting the imbalanced microbiota to healthy microbiota,thus improving the prognosis of caries patients.
