Background:Micro RNAs(miRNAs)have become one of the research hotspots in recent years,because they can participate in the regulation of gene expression.It has been reported that micro RNA-513c(miR-513c)can affect the proliferation,migration and invasion of neuroblastoma,glioma and other malignant tumors by regulating related target genes.Therefore,based on the existing theoretical basis,the expression and function of miR-513 c and Cul4 A were studied in HCC tissues.Objectives:This paper intends to study the expression of miR-513 c and Cul4 A in hepatocel-lular carcinoma(HCC)tissues and adjacent non-cancer tissues,and further study their mutual regulation.Methods:1.Specimens of 85 pairs of HCC tissue and adjacent non-carcinoma tissue(2cm from the edge of carcinoma tissue)that underwent surgical resection of HCC lesions were collected.2.The target genes and HCC-related genes of miR-513 c were predicted on different bioinformatics websites,the relationship between miR-513 c and Cul4 A was evaluated,and the targeted binding sites were verified by RIP and double luciferase reporter gene experiments.3.The expression levels of miR-513 c and Cul4 A in 85 pairs of HCC tissues,paracancer non-cancer tissues and constructed Hep G2 cell models were detected,and the relationship was found.4.The proliferation,invasion,migration and apoptosis of Hep G2 cells were evaluated by cc K-8,flow cytometry and Transwell assay.Results:1.According to relevant data,Cul4 A was analyzed and verified as the direct target gene of miR-513 c.2.The expression level of miR-513 c in HCC tissues was decreased,while the expression level of Cul4 A was significantly increased.3.Using Hep G2 as cell model and transfection group design,miR-513 c negatively targeted Cul4 A was observed.4.Overexpression miR-513 c targets Cul4 A to inhibit the proliferation,invasion and migration of HCC cells and promote apoptosis.Conclusion:1.The downstream target gene of miR-513 c is Cul4 A.2.miR-513 c negatively targets Cul4 A,Overexpression miR-513 c may inhibit the proliferation,invasion and migration of HCC cells and promote apoptosis.3.Decreased miR-513 c expression may be a potential adverse prognostic factor for HCC patients.It is expected to be a new potential target for HCC gene therapy.
