| Background and purpose:Osteosarcoma is one of the most common malignancies in orthopedics,and chemical resistance often results in a poor prognosis for patients with osteosarcoma.m RNA has the advantage of being uniquely targeted as a target for drug therapy.By developing a risk model for osteosarcoma prognostic genes,the tumor prognostic factor FKBP11 was found to have a strong correlation with osteosarcoma.This paper is dedicated to investigate the effect of FKBP11 on the proliferation,invasion and migration of osteosarcoma and apoptosis during the development of osteosarcoma,and to provide a new idea for the treatment of osteosarcoma.Method:Data were obtained from databases such as GTEx,and differential analysis of expression data was performed using gene set enrichment analysis.The set of associated genes with significantly different expression was obtained,and their enrichment score for this gene set was performed.Then,two differential genes associated with prognosis were screened by univariate Cox regression analysis,least absolute contraction and selection operator(lasso)Cox regression analysis and multifactorial COX regression analysis,and a risk assessment model for tumor prognosis-related genes was constructed.QPCR was used to detect the expression of FKBP11 in osteosarcoma cells;and the expression of FKBP11 in osteosarcoma tissues was detected by immunohistochemical staining.The expression of FKBP11 was knocked down in osteosarcoma cells,and the proliferation,invasion and migration ability of osteosarcoma cells were observed using cell proliferation,Transwell and colony formation assays,and the apoptosis of osteosarcoma cells was measured using Annexin V-FITC / PI kit.Finally,the expression of FKBP11 was examined by Western blotting,and the effect of knockdown of FKBP11 on invasion and migration-related proteins E-Cadherin,N-Cadherin and Vimentin,apoptosis-related proteins Cleaved Caspase3,and MAPK signaling pathway proteins p-MEK and p-ERK was examined.Results:1、 Screening and enrichment of osteosarcoma-related genes;2、 Construction of a tumor prognosis-related genetic risk assessment model;3、 FKBP11 and BNIP3 are associated with the degree of risk of osteosarcoma;4、 FKBP11 expression is significantly increased in osteosarcoma;5、 Knockdown of FKBP11 reduces the ability of osteosarcoma cells to proliferate,invade and migrate;6、 Knockdown of FKBP11 reduces MAPK pathway protein expression.Conclusion:FK506-related protein 11 is associated with with the prognosis of osteosarcoma.FKBP11 is highly expressed in osteosarcoma,and FKBP11 can affect the proliferation,invasion and migration and apoptosis of osteosarcoma cells by mediating MAPK signaling pathway,and FKBP11 is a potential target for osteosarcoma treatment,which is important for the treatment of osteosarcoma. |
