Objective:To study the expression of Smo and Gli1 proteins in eyelid basal cell carcinoma tissues,analyze the relationship between the expression of Smo and Gli1 and the clinical characteristics of eyelid basal cell carcinoma,and explore the correlation of Smo and Gli1 proteins and their clinical significance in eyelid basal cell carcinoma.Methods:A total of 32 cases of eyelid basal cell carcinoma(BCC)who were surgically resected and pathologically diagnosed in the department of Ophthalmology of the Second Affiliated Hospital of Nanchang University from 2018 to 2021 were selected as the experimental group and the control group,respectively.There were 11 males and 21 females;18 cases were aged 65 years and above,and 14 cases were under 65 years old;15 cases invaded the orbicularis oculi muscle layer,and 17 cases invaded the orbital diaphragm and orbital fat layer;the tumor diameter was greater than or equal to 12 cases were 2 cm,20 cases were less than 2 cm;17 cases had tumor course of 2 years or more,and 15 cases had tumor course of less than 2 years.Immunohistochemical staining was used to detect the expression of Smo protein and Gli1 protein in the two groups.Image J graphic analysis software was used to analyze the optical density of the immunohistochemically stained paraffin sections,and the average optical density of positive particles in the sections was calculated.SPSS 25.0software system was used for statistical analysis,and P < 0.05 was considered statistically significant.Results:1.(1)The positive expression rate of Smo in eyelid basal cell carcinoma tissues was 65.68%(21/32),and 21.88%(7/32)in the adjacent normal tissues.The average optical density of the experimental group was 0.264±0.063,and that of the control group was 0.192±0.042,The P value of Mann-Whitney test was 0.000(P <0.05).The optical density of Smo protein in eyelid basal cell carcinoma was higher than that in adjacent normal tissues,the difference was statistically significant.(2)The positive expression rate of Gli1 in eyelid basal cell carcinoma tissues was 71.88%(23/32),and25.00%(8/32)in the adjacent normal tissues.The average optical density of the experimental group was 0.271±0.067,and that of the control group was 0.196±0.041.Mann-whitney test showed that the P value was 0.000(P <0.05).The optical density of Gli1 protein in eyelid basal cell carcinoma was higher than that in the adjacent normal tissues,the difference was statistically significant.2.(1)The average optical density of Smo was 0.229±0.054 in the tumor group of invasion orbicularis muscle carcinoma,and 0.296±0.054 in the tumor group of invasion orbital diaphragm and orbital adipostasis carcinoma.The P value was 0.000 by Mann-Whitney test(P <0.05),and the difference was statistically significant.The average optical density values expressed by Smo in the groups of gender,age,tumor diameter and tumor course were all greater than 0.05 by Mann-Whitney test,and the difference was not statistically significant.(2)The average optical density of Gli1 was0.225±0.058 in the tumor group of invasion orbicularis muscle carcinoma,and0.311±0.047 in the tumor group of invasion orbital diaphragm and orbital adipocytes carcinoma.The P value was 0.000 by Mann-Whitney test(P<0.05),and the difference was statistically significant.The average optical density of Gli1 expressed in the groups of gender,age,tumor diameter and tumor course was all greater than 0.05 by Mann-Whitney test,and the difference was not statistically significant.3.Spearman correlation analysis showed that the expressions of Smo and Gli1 proteins were positively correlated in eyelid basal cell carcinoma tissues(r=0.735,P<0.05)Conclusions:1.The expression of Smo and Gli1 protein in eyelid basal cell carcinoma tissues was higher than that in the control group,and the difference was statistically significant.2.The expression of Smo and Gli1 protein in eyelid basal cell carcinoma was correlated with the tissue level of tumor invasion,but was not related to gender,age,tumor diameter and tumor course.3.the expression of Smo and Gli1 protein in eyelid basal cell carcinoma is positively correlated,which may be synergistically involved in the occurrence and development of eyelid basal cell carcinoma.
