Objective:Fibroblast activation protein(FAP)is a transmembrane serine protease,which is generally expressed on activated fibroblasts in the tumor microenvironment(TME),and involved in the process of tumor occurrence and development,proliferation,invasion,angiogenesis and so on.However,in a number of relevant clinical studies,it is found that its impact on tumor prognosis is not completely consistent,and there is a certain degree of controversy.Therefore,in order to further explore the correlation between FAP and tumor prognosis,this paper uses the method of meta-analysis to analyze the relevant data in order to solve this problem.Methods:The relevant literatures of CNKI,Pub Med,EMBASE and Cochrane library databases were searched.According to the inclusion and exclusion criteria,the literatures were selected,and the data of six outcome indexes such as overall survival(OS),disease-free survival(DFS),lymph node metastasis,tumor infiltration,TNM staging and pathological grade were collected.Finally,processing and analyzing the data software stata16.0.Results:(1)In the overall analysis,the high expression of FAP was negatively correlated with the overall survival and disease-free survival of solid tumors,and increased the risk of lymph node metastasis,tumor infiltration and advanced tumor development,but it had no correlation with the pathological grade.(2)In subgroup analysis based on solid tumor types,it was found that the effect of FAP overexpression on OS and DFS in esophageal cancer group was consistent with the overall analysis.The decrease of OS in lung cancer,pancreatic cancer and gastric cancer was related to the high expression of FAP.It was also found that the high expression of FAP in gastric cancer group was positively correlated with lymph node metastasis,tumor invasion,and the risk of progression to late stage cancer,but only in the analysis of tumor invasion,and there was no correlation in data analysis of pancreatic cancer.(3)In the subgroup analysis based on the positive expression site of FAP,the effect of high expression of FAP in stroma on OS,DFS,lymph node metastasis and tumor invasion is consistent with the overall analysis results,while the high expression of FAP in tumor tissue has no correlation with OS,lymph node metastasis and tumor invasion,but the high expression of FAP in tumor tissue is more obvious in advanced tumors,but there is no correlation in tumor stroma.Conclusion:The high expression of FDP is likely to indicate the poor prognosis of solid tumors.In particular,compared with tumor tissues,the high expression of FDP in tumor stroma is closely related to the poor prognosis of tumor patients,which is worthy of further study. |
