Objective:The implementation of RNA sequencing to determine circular RNA(circ RNA)expression spectrum in autologous arteriovenous fistula(AVF)tissues of uremia patients.To investigate the possible relationship between circ RNA and the progression of AVF vascular stenosis.Methods:4 cases of healthy control group: from February 2017 to May first affiliated hospital of nanchang university renal medicine in 54 cases of uremic patients were diagnosed with randomly selected from 4 cases out of vascular stenosis and blood vessel disease patients and control group in 4 cases,nanchang university from February 2017 to May,the first affiliated hospital urology department in 54 cases of uremic patients were diagnosed with randomly selected from 4 patients with diagnosis of AVF narrow.Diagnostic criteria for AVF stenosis: AVF service life of more than 3 months for arteriovenous internal fistula;clear imaging results of upper limb vessels such as ultrasound;AVF blood flow during hemodialysis was <200m L/min;The degree of stenosis is greater than 50%,excluding obvious thrombosis.Then total RNA samples of the experimental group and control group were extracted to detect the difference of circ RNA expression profile and the differentially expressed circ RNA was selected(p < 0.05,Differential expression ≥2times).Cluster analysis,gene function analysis and KEGGpathway analysis were performed.Real-time quantitative PCR(q RT-PCR)was used to verify the results of RNA sequencing.Results:Among 17620 circ RNA transcripts detected by RNA sequencing,208 circrnas were significantly differentially expressed between the experimental group and the control group(P < 0.05,Differential expression ≥2 times),among which 92 were up-regulated and 116 were down-regulated.The 8 circrnas were subsequently verified by q RT-PCR,Only hsacirc_015036 was consistent with the sequencing resultsConclusion:It is true that circ RNA is different-expressed in the stenosis and non-stenosis tissues of autogenous arteriovenous fistula in uremia patients,hsacirc_015036 and Focal adhesion signaling pathway may play an important role in the occurrence and development of AVF venous stenosis in uremia patients.
