| In the long historical development process of the Chinese nation,due to the difference in the growth,planting and medication habits of traditional Chinese medicines in the north and south,many Traditional Chinese medicines have the same name foreign body.Modern Chinese medicine has done more in-depth research on the same family of Chinese medicines with similar chemical components in appearance.More and more research results show that there are great differences in the characteristics,clinical efficacy and safety of the same family of North and South Chinese medicines,which are related to the chemical components contained in them.The types and proportions of compounds contained are the material basis for the differences between northern and southern Chinese medicines.The literature reports that there are safety problems in a variety of traditional Chinese medicine volatile oils,including hepatotoxicity,genotoxicity,cardiotoxicity,pulmonary toxicity,etc.,of which liver safety problems account for the highest proportion.The liver is the largest metabolic organ in the human body,and hepatotoxicity problems can cause drug metabolism disorders and liver damage.As a widely used traditional Chinese medicine,Schisandra chinensisand Bupleurum chinensis contain volatile oils that are one of the main active ingredients.There have been many reports of hepatotoxicity of volatile oil in Bupleurum chinensis,but the basic research on substances is still blank;Schisandra chinensis has a unique advantage in the treatment of liver disease,and as one of the main components of Schisandra chinensis oil,the safety issue is worth exploring.In this project,the safety assessment of these two northern and southern medicinal herbs focused on the liver and explored the impact on the safety of LO2 hepatocytes.Drug safety is closely related to the basis of the substances contained,so this topic compares the safety differences between extraction and safety evaluation of Schisandrae Sphenantherae and Schisandrae chinensis oil/Bupleurum scorzonerifolium Willd and Bupleurum chinensis DC oil on LO2 liver cells based on the analysis of the substances of the compounds contained.In terms of safety evaluation,LO2 liver cells were cultured in vitro,and the effects of Schisandrae Sphenantherae and Schisandrae chinensis oil/Bupleurum scorzonerifolium Willd and Bupleurum chinensis DC oil on the morphology,number,proliferation and apoptosis of LO2 liver cells were observed and detected by cell morphology,CCK-8 detection and flow cytometry.According to the results of Schisandrae Sphenantherae oil under the 4000μg/m L concentration on LO2 liver still no obvious inhibition of cell proliferation,cell morphology and cell apoptosis rate had no significant change,IC50cannot be calculated.Schisandrae chinensis oil could inhibit cell proliferation and increase apoptosis rate in a concentration-dependent manner(*P<0.05)with IC50of(1050±89.81)μg/m L.The safety of Schisandrae Sphenantherae oil on LO2 liver cells was significantly different from that of Schisandra chinensis oil,and the safety dose of Schisandrae Sphenantherae oil was much higher than that of Schisandra chinensis oil.Otherwise,the IC50of Bupleurum scorzonerifolium Willd oil is(331.42±12.43)μg/m L.The IC50of Bupleurum chinense DC oil is(809.17±32.64)μg/m L.In addition,Western Blotting was used to further explore the potential mechanism of the different safety of Bupleurum scorzonerifolium Willd and Bupleurum chinense DC oil on liver cells.The results showed that Bupleurum scorzonerifolium Willd and Bupleurum chinense DC oil inhibited the proliferation of LO2 hepatocytes by affecting the expression of Bcl-2and Bax proteins.At the same time,combined with the results of the chemical composition and difference markers of the two oils,it was found that the aldehydes and alkenes contained in the oils may be the potential material basis for the difference in liver function safety of while the alkenes and phenols contained in the oils may be the potential material basis for the difference in liver function safety of Schisandrae Sphenantheraeand Schisandrae chinensis oil,while the alkenes and phenols contained in the oils may be the potential material basis for the difference in liver function safety of Bupleurum scorzonerifolium Willdand Bupleurum chinensis DC oil.The extraction of traditional Chinese medicine volatile oil safety control is the hot topic of research,is also restricting Chinese medicine volatile oil of clinical medicine and traditional Chinese medicine volatile oil formulation development an important bottleneck.In this paper,the optimal extraction conditions of two common volatile oil extraction methods were explored.The safe dose of Schisandra oil and Bupleurum oil on LO2 hepatocytes was determined.By comparing thechisandrae Sphenantheraeand Schisandrae chinensis oil/Bupleurum scorzonerifolium Willdand Bupleurum chinensis DC oil,analyzing differences between the south and north Chinese medicine volatile oil security potential compounds,the extraction of volatile oil preparations for traditional Chinese medicine,the research,development and characteristic provides the basis for the establishment of safety and quality control method,the distinction between traditional Chinese medicine from north to south(TCM)clinical use has played a warning role. |
