Using cerebral ischemia reperfusion injury(CIRI)and vascular dementia(VD)rat models of ischemic cerebral disease(ICD)as the research object,to study the protective effect and potential mechanism of ginsenoside Rg3(GRg3)on ICD rats.In the ICD rat model of CIRI,GRg3 was administered prophylactically to CIRI rats,the protective effect of GRg3 in CIRI rats was evaluated by Morris water maze test and antioxidant capacity assay,and the ultrastructure of brain mitochondria was examined by transmission electron microscopy.Metabolomic analysis based on gas chromatography-mass spectrometry(GC-MS)method and apoptosis analysis based on western blot to evaluate the potential mechanism of GRg3 protection in CIRI rats.The results showed that prophylactic administration of GRg3 can improve the cognitive ability of CIRI rats,protect the brain cell and mitochondrial ultrastructure of CIRI rats,and reduce the oxidation level of brain and blood in CIRI rats.The potential mechanism of its protective effect is related to the regulation of mitochondrial dysfunction,including regulation of mitochondrial metabolic disturbances and anti-mitochondrial pathways to apoptosis.In the ICD rat model of VD,GRg3 was administered therapeutically to VD rats,and the protective effects on VD rats were evaluated by Morris water maze test,transaminases and blood lipids,and evaluation of changes in fecal microbiota in VD rats based on 16 S r RNA gene detection technology.Metabolomic analysis based on GC-MS method and cellular inflammation analysis based on western blot to evaluate the potential mechanism of GRg3 protection in VD rats.The results showed that therapeutic administration of GRg3 could improve the cognitive ability of VD rats,protect the cellular structure of the hippocampus in VD rats,improve the fatty lesions of liver cells in VD rats,and reduce the four levels of blood transaminases and blood lipids in VD rats.The potential mechanism of protection is related to increasing the abundance of probiotics in the feces of VD rats,inhibiting the JNK and P38 MAPK inflammatory signaling pathways in the liver-brain axis cells of VD rats,and improving the abnormal metabolism of the gut-liver-brain axis in VD rats.This study showed that GRg3 had a protective effect on ICD rats.At the same time,the experimental data of this study also provide data support for the clinical study of GRg3 in the treatment of ICD.
