Objective:To study the changing characteristics of type 2 innate lymphoid cell(ILC2),ILC2-related characteristic nuclear transcription factors and inflammatory cytokine in the peripheral blood of healthy children and children with mycoplasma pneumoniae pneumonia,for exploring whether ILC2 is involved in the immune response to mycoplasma pneumoniae pneumonia.Methods:25 cases of children with MPP hospitalized in the Zun Yi Maternal and child health hospital from March 2021 to November 2021 were randomly selected as MPP group.In the same period,25 cases of health children in Children,s Health Care Department were randomly selected as normal control group.Basic clinical data of children with MPP and healthy children were collected,and fasting venous blood was collected.The proportion of ILC2 in peripheral blood mononuclear cell,the proportion of CD4~+T cells and CD8~+T cells in peripheral blood were determined by flow cytometry.Enzyme-linked immunosorbent assay was used to detect the concentrations of interleukin-33,growth stimulation expressed gene 2,interleukin-4,interleukin-13,interferon-γand immunoglobulin G.Quantitative real-time polymerase chain reation was used to detect the transcriptional levelof retinoid acid receptor related orphan receptorαand GATA-binding factor 3.All of the data were subjected to statistical analysis.Results:(1)There were no significant differences in age,sex,leukocyte number and lymphocyte number in the NC and MPP group(P>0.05).(2)The proportion of ILC2and the level of ILC2-related nuclear transcription factors retinoid acid receptor related orphan receptorα,GATA-binding factor 3 in the MPP group were significantly higher than NC group,the difference was statistically significant(P<0.05).(3)The level of interleukin-4,interleukin-13,interferon-γ,interleukin-33,growth stimulation expressed gene 2 in the plasma of patients of MPP group was higher than the NC group,the difference was statistically significant(P<0.05),ILC2 and interleukin-4,interleukin-13 and growth stimulation expressed gene 2,interleukin-33and ILC2 were positively correlated(P<0.05),ILC2 and interferon-γwere inversely correlated(P<0.05).(4)Compared with the normal control group,the proportion of CD8~+T cells and the level of immunoglobulin G was significantly increased,the proportion of CD4~+T cells was decreased,thedifference was statistically significant(P<0.05),but them were not associated with ILC2(P>0.05).Conclusion:(1)ILC2 is involved in the pathogenesis of mycoplasma pneumoniae pneumonia,secreting interleukin-4 and interleukin-13 affects the immune status of the children with mycoplasma pneumoniae pneumonia,mediates the Th2 immune responses and promotes the development of lung inflammation.(2)Upon mycoplasma pneumoniae infection,IL-33 promotes ILC2 activation.
