Objective:Sarcandra glabra(Thunb.)Nakai has the effect of clearing heat and activating blood,and it is commonly used to treat pelvic inflammatory disease(PID)and the sequelae of pelvic inflammatory disease in the folk.Therefore,the purpose of this study is to explore the effect and the preliminary mechanism of Sarcandra glabra on PID,so as to lay an experimental foundation for its clinical application.Methods:(1)The experimental models of xylene-induced ear edema,acetic acid-induced vascular permeability and granuloma test in mice were used to observe the inflammatory effect of Sarcandra glabra.(2)The experimental models of hot plate test,acetic-induced writhing and formalin-induced liking test were used to investigate the analgesic effect of Sarcandra glabra.(3)PID rat model was duplicated by the mixed bacteria of Escherichia coli and Staphylococcus aureus combined with mechanical injury.All rats were randomly divided into 6 groups:blank control group,model group,positive control group,high-dose group of Sarcandra glabra(7g/kg),middle-dose group of Sarcandra glabra(3.5g/kg)and low-dose group of Sarcandra glabra(1.75g/kg).Experimental drugs were given intragastric administration for 14 days respectively after the PID rat model was successfully established.And then the pathological changes of uterus,ovaries and pelvic tissue of PID rats in each group were observed by naked eyes and Hematoxylin-eosin(HE)staining sections.The coefficients of uterus,fallopian tubes,ovaries,spleen and thymus were calculated.The contents of PGE2,histamine,5-HT and FGF-2 in serum were detected by enzyme-linked immunosorbent assay and fluorescence spectrophotometry respectively.Results:(1)7g/kg and 3.5g/kg dose groups of Sarcandra glabra could significantly inhibit ear edema induced by xylene(P<0.05),inhibit the increase of vascular permeability induced by acetic acid(P<0.05),and reduce the weight of granuloma(P<0.05).(2)7g/kg,3.5g/kg and 1.75g/kg dose groups of Sarcandra glabra had no significant effect on the pain threshold of mice induced by hot plate stimulation(P>0.05).7g/kg dose group of Sarcandra glabra could prolong the latency time of pain induced by acetic acid in mice(P<0.05),while 7g/kg and 3.5g/kg dose groups of Sarcandra glabra could reduce writhing induced by acetic acid in mice(P<0.05)and reduce the pain response induced by formalin in rats(P<0.05).(3)7g/kg,3.5g/kg and 1.75g/kg dose groups of Sarcandra glabra could significantly reduce the pathological changes in PID rats such as congestion,edema and adhesion of pelvic tissue.7g/kg and 3.5g/kg dose groups of Sarcandra glabra could significantly decreased the coefficient of uterus(P<0.05),but the three groups had no significant effect on the coefficient of fallopian tubes(P>0.05).7g/kg dose group of Sarcandra glabra could significantly decreased the coefficient of ovaries(P<0.05).7g/kg dose group of Sarcandra glabra could significantly increase the coefficients of spleen and thymus(P<0.05).(4)The content of PGE2 in serum was significantly decreased in 7g/kg and 1.75g/kg dose groups of Sarcandra glabra(P<0.05).The content of histamine in serum was significantly decreased in 3.5g/kg and 1.75g/kg dose groups of Sarcandra glabra(P<0.05).And the content of 5-HT in serum was significantly decreased in 7g/kg dose group of Sarcandra glabra(P<0.05).(5)The content of FGF-2 in serum was significantly decreased in 7g/kg and 3.5g/kg dose groups of Sarcandra glabra(P<0.05).Conclusions:Sarcandra glabra has significant anti-inflammatory effect and certain analgesic activity.It can improve the pathological changes of pelvic inflammatory disease,and its mechanism may be related to the decrease of serum PGE2,histamine,5-HT,and FGF-2 contents. |
