Objective:This study investigates the modern medical mechanism of action of Chinese medicine urinary bladder matrix(UBM)in repairing skin injury wounds based on a combination of Chinese and Western medicine,and compares it with artificial type I collagen scaffolds(PEL)to add new scientific evidence to the medicinal value of Chinese medicine "flesh and blood products" and promote the development of Chinese animal medicine.Methods:Material experiment:(1)Scanning electron microscope was used to observe the morphology of UBM and PEL materials;(2)The soluble components of scaffolds were extracted by soaking in urea-containing buffer and then subjected to SDS-PAGE experiment to observe the protein distribution of the components.In vitro experiments:(1)Scanning electron microscope and immunofluorescence staining were used to investigate the adhesion and growth of cells on scaffolds;(2)Scratch and Transwell assay to detect the effect of the extract on the migration movement of cells;(3)Flow cytometry was used to detect the effect of the extract on the polarization of macrophages.In vivo experiments:(1)In vivo implantation experiments in mice to explore the effect of endogenous cells on the growth of the scaffold and the surrounding microenvironment;(2)H&E staining to observe the infiltration of cells in the material;(3)Anti-ER-TR7,F4/80,α-SMA immunohistochemical staining and F4/80,CD86,CD206 immunofluorescence co-staining to assess the infiltrating cell types and activation;(4)anti-TGF-β1,VEGF,MMP-9 and Col I immunohistochemical staining was used to evaluate the microenvironment and scaffold remodeling after implantation.Results:1.One side of UBM is a dense basement membrane structure,and the other side is a porous lamina propria containing a fibrous structure.Both the upper and lower sides of the PEL have a three-dimensional porous structure.The extract of UBM contains some non-type I collagen components.2.UBM can promote the adhesion and growth of fibroblasts planted on its surface in a serumindependent manner.The extract also significantly promoted cell adhesion,proliferation and migration,and also significantly promoted the polarization of macrophages to M2 type.3.UBM also showed significantly enhanced rapid endogenous cell ingrowth activity after implantation in vivo,while creating a microenvironment rich in TGF-β1 and VEGF,inducing cell activation,generating type I collagen,and promoting scaffold remodeling,thereby promoting in situ tissue regeneration.Conclusion:1.UBM has more significant biological activities to promote cell adhesion,proliferation,migration and activation,induce pro-regeneration and pro-remodeling of the microenvironment,and promote the polarization of macrophages to M2 type,thereby promoting the constructive remodeling of scaffolds.2.Some of the advantageous functions of Chinese medicine "flesh and blood products" are irreplaceable by synthetic substances.3.More modern medical mechanisms of action of Chinese animal medicine should be explored to increase their scientific basis,so as to advance the development of Traditional Chinese Medicine. |
