Effect Observation And Mechanism Exploration Of Shaogan Dingxiao Granules In Treating Children Asthma With Yin Deficiency Of Liver-heat And Wind-moving Syndrome

Objective: To observe the clinical efficacy of Shaogan Dingxiao Granules in treating children asthma with Yin Deficiency of Liver-Heat and Wind-Moving Syndrome,and explore the mechanism of its network pharmacology.Methods: 1.80 cases of children were divided into two groups,the control group(40 cases)was given Budesonide combined with Compound Ipratropium Bromide Solution for Inhalation,and the treatment group(40 cases)was given Shaogan Dingxiao Granules orally,both were treated for 7 days.Record the patients’ condition before and after treatment.2.Obtain drug active ingredients and related targets through TCMSP,BATMAN and Swiss Target Prediction,and obtain related targets of childhood asthma from Gene Cards.Draw Drug-Disease Venn diagrams to derive common targets.The visualization network of Drug-Active Ingredient-Disease-Target was constructed with the help of Cytoscape software,the PPI network was constructed using the STRING database,and the GO and KEGG enrichment analysis results were obtained and visualized in the DAVID database.Results: 1.After comparison,there was no statistical difference in the baseline data of the two groups of children,and they were comparable(P>0.05).2.A total of 5 cases dropped out in this study(2 cases in the treatment group and 3 cases in the control group),and 75 cases were finally completed(38 cases in the treatment group and 37 cases in the control group).After treatment,the total syndrome score,the main symptom score and the secondary symptom score in the two groups were significantly decreased,with statistical difference(P < 0.05).3.Efficacy evaluation: There was no statistical difference in the total effective rate between the two groups(P>0.05),the effective rate in the treatment group was 97%,and the effective rate in the control group was 95%,but the number of markedly effective patients in the treatment group was more than that in the control group.4.During the clinical observation period,there were no adverse events,and the safety evaluation was Grade One.5.A total of 149 active ingredients of traditional Chinese medicine,1156 targets related to active ingredients,500 targets related to childhood asthma,and 157 common targets were screened out.The key active ingredients of this recipe for asthma are stigmasterol,beta-sitosterol,quercetin,baicalein,and betulinic acid,and the key targets are TNF,ALB,AKT1,IL-1β,and VEGFA.This study involved a variety of biological processes such as drug response,apoptosis process,vasoconstriction,and multiple signaling pathways such as HIF-1,calcium signaling pathway,and FOXO.Conclusions: 1.Shaogan Dingxiao Granules have a significant curative effect on children asthma with Yin Deficiency and Liver-Heat and Wind-Moving Syndrome,and are significantly better than the aerosol control group in the improvement of secondary symptoms.2.During the clinical observation period,Shaogan Dingxiao Granules are safe in the treatment of children asthma with Yin Deficiency and Liver-Heat and Wind-Moving Syndrome.3.This recipe may inhibit the expression of TNF,AKT1,IL-1β,VEGFA and other proteins and HIF-1,FOXO and other signaling pathways through compounds such as stigmasterol,beta-sitosterol,quercetin,baicalein and betulinic acid,thereby exerting anti-inflammatory,Antioxidant and improve hypoxia effects.
