| Objective: In this paper,by studying the expression of Tim-4 in primary liver cancer(PHC)pathological cancer tissues,adjacent tissues and peripheral blood mononuclear cells,as well as the concentration of s Tim-4 in plasma,the role of Tim-4 in PHC was clarified.Expression levels and clinical value of Tim-4 in PHC disease.Methods:1.Collect 30 pairs of cancer tissue and corresponding paracancerous tissue pathological sections of patients with PHC diagnosed by pathology in Guilin People’s Hospital and Guangxi Zhuang Autonomous Region People’s Hospital,and 23 liver tissue sections from non-PHC patients.pathological data.Immunohistochemical experiments were used to detect the expression level of Tim-4 in PHC cancer tissue,adjacent tissue and liver tissue of non-PHC patients,and the correlation between its expression level and clinicopathological data.2.Collect the peripheral blood of 29 patients newly diagnosed with PHC and 30 normal controls during the same period.Flow cytometry was used to detect the expression ratio of monocyte subsets and the expression levels of Tim-4 in monocytes and monocyte subsets;The difference of plasma s Tim-4concentration between PHC group and normal control group was analyzed.At the same time,the clinical data of AFP,ALB,ALP,GGT,AST,ALT,hepatitis history and other clinical data of all patients were collected.The correlation between Tim-4 expression and clinicopathological data was analyzed and discussed.The diagnostic efficacy of Tim-4 for PHC was analyzed by ROC curve,and the predictive value of s Tim-4 for PHC was analyzed by logistic regression.Results:1.Tim-4 is lowly expressed in PHC cancer tissues,and highly expressed in adjacent and non-PHC liver tissues.The expression level of Tim-4 in cancer tissues is related to age,gender,tumor differentiation,tumor size,tumor number,history of hepatitis B,AFP and other clinical data were not statistically different.2.It was found by flow cytometry that the expression ratio of intermediate monocyte subsets in the PHC group was 8.3(4.8,11.85)%higher than that of the normal control group,3.35(2.58,6.65)%,P < 0.05;The expression ratio of monocytes in the PHC group was 61(45.8,74.45)%and 69.3(45.7,80.68)% in the normal control group(P > 0.05);the non-classical monocyte subset was 14.3(6.65,24.45)% and the control group 19.35(8.63,31.45)% also had no statistical difference(P > 0.05).It is suggested that intermediate monocytes may be involved in the progression of hepatocellular carcinoma.3.The expression of Tim-4 in monocytes of PHC patients was 9.1 %(3.65,14.8)and the control group was 4.7 %(2.05,9.4),there was a statistical difference(P < 0.05),and it was mainly expressed in the subtype of intermediate monocytes.The expression levels of the two groups were significantly different(P < 0.05)compared with 73.85 %(57.7,85.3)of the control group.It shows that the expression of Tim-4 is closely related to the intermediate monocyte subset,and in tumor diseases,the intermediate monocyte subset usually shows an increasing trend,suggesting that the increase of Tim-4 may be related to tumor progression..4.Monocyte Tim-4 and peripheral blood s Tim-4 were positively correlated with AFP,ALT,AST,GGT,ALP,and negatively correlated with ALB,suggesting that the expression level of Tim-4 is related to hepatocyte injury and liver anabolic function It can be used as a potential indicator for liver function evaluation.5.The plasma s Tim-4 concentration in the group with a history of hepatitis was 11.62 ng/m L(6.83,18.75),which was higher than that in the group without a history of hepatitis B,6.10 ng/m L(3.21,8.03),P < 0.05,indicating that s Tim-4 may be involved in the process of PHC.Inflammation-cancer transformation.6.The ROC curve shows that s Tim-4 combined with AFP can improve the diagnostic performance of PHC.Logistic regression analysis shows that patients with increased s Tim-4 have a 2.29-fold higher risk of cancer.Conclusion(s):Tim-4 is lowly expressed in PHC cancer tissues,and its expression level has no statistically significant correlation with the clinical characteristics of tumors.The proportion of intermediate monocyte subsets is significantly increased in PHC disease,and Tim-4 is highly expressed in PHC monocytes.The level was positively correlated with AFP,TBIL,GGT,ALP,AST,and ALT of PHC,and negatively correlated with its ALB and PA,suggesting that Tim-4 can be used to evaluate the damage degree and synthesis ability of hepatocytes.The expression level of Tim-4 was highly expressed in the hepatitis virus infection group,suggesting that Tim-4 may be involved in the inflammatory-cancer transformation of the liver.In this study,the combined detection of s Tim-4 and AFP was used for the first time,which can improve the diagnostic performance of PHC patients,and patients with elevated s Tim-4 had a 2.29-fold increased risk of disease. |
